Objective:This experiment in GTW of follicular development rats model as the research object,from CyclinD2, PCNA, Bax,Bcl-2 perspective to explore the xinjiaguishenwan treatment of follicular developmental disorder, for clinical provides new ideas and methods in the future.Method:with 54 female SD rats, random 9 as blank control group, only the remaining 45 with 75mg/(kg.d) GTW lavage 30 days, copy of follicular development rats model, evaluation after the success of the building, will be randomly divided into 5 groups, namely:model control group, high dose group, chinese medicine dose group, low dose group, the western medicine group.The blank control group and model control group were filled daily with distilled water; the western medicine group were treated with estrogen and progestin sequential through intra-gastric administration;three different dosages of herbal compound xinjiaguishenwan group were given different dose of chinese medicine in the same way. All groups were treated for 2 weeks ontinuously. Vaginal exfoliated cells smears were operated during the treatment to observe oestrous cycle of the rats.24 hours after the last treatment, all the rats were killed after taken femoral artery blood. Then the rats’abdominal cavity were opened and both ovaries were taken out. Detected the expression of cyclinD2mRNA, PCNAmRNA, BaxmRNA and Bcl-2mRNA with RT-PCR.RESULTS:GTW 75mg/(kg-d) gavage for 30 days can result in follicular development disorder of the child-bearing period SD rats. model control group rats showed the disorders of estrous cycle, the decreasing of ovarian indexes, mRNA levels of cyclinD2, PCNA, Bax/Bcl - 2 ratio in rat ovary,The above indicators had significant difference compared with the blank control group (P<0.05 or P<0.01).After treatment with Xinjiaguishenwan for two weeks, the model rats had obvious improvement in their general conditions. The high dose group and the western medicine group showed normal or close to the normal estrous cycles, the increasing of ovarian indexes, mRNA levels of cyclinD2 in rat ovary, The above indicators had significant difference compared with the model control group (P<0.05), The high dose group showed mRNA levels of PCNA, Bax/Bcl-2 ratio in rat ovary, The above indicators had significant difference compared with the blank control group (P<0.05).Conclusion:xinjiaguishenwan can improve GTW of follicular development rats model hormone secretion from ovarian function, at the same time raise mRNA levels of cyclinD2, PCNA in rat ovary, Bax/Bcl-2 gene expression reduction ratio, Therefore resuming the development of follicle, renovate the damaged function of the ovaries.
