Expression of CyclinD2 and IGFBP2 in HL and ALCL and its significanceObjective:This study aims to detect the expression of cyclinD2 and IGFBP2 in HL and ALCL at thelevel of protein and to investigate the roles of the two genes in the pathogenesis of HLand ALCL.Methods:HE staining, the monoclonal and polyclonal antibodies CD30(BerH2), CD15(C3D-1),CD45RO(UCHL1) andCD20(L26) were used for selecting the cases of HL and ALCLfrom 288 cases of malignant lymphomas. The clarified paraffin embeded tissue,comprising cases of HL and cases of ALCL were finally subjected to immuno-histochemical staining to analyze the expression of cyclinD2 and IGFBP2.Results:1. 26 cases of HL and 19 cases of ALCL were confirmed by HE staining and immunohistochemically by CD30, CD15, CD20, CD45RO staining.2. The positive rate of cyclinD2 expression in 26 cases of HL was 92% (24/26), among them 15 cases were stained +++, 7 cases were stained ++, 2 cases were stained +, the other 2 cases were negatively stained. The positive rate of it in ALCL cases was 36.8% (7/19), including 1 case strongly stained +++, 2 cases stained ++, 4 case stained +, but 12 other cases negatively stained. There presented a statistic difference between HL and ALCL groups (p=0.000<0.05) for the positive rate of cyclin D2.3. The positive rate of IGFBP2 expression in HL was 23/26 (88.5%), including 14 cases strongly stained +++, and 9 cases stained ++. Out of 19 cases of ALCL, only 2 cases were stained ++, 4 cases were stained +, the other 13 cases were negatively stained, The positive rate of it in ALCL cases was 31.6%. There presented a statistic difference between HL and ALCL groups (p=0.000<0.05) for the positive rate of IGFBP2 expression.4. 2 cases showed to be a mixed type combined with large tumor cells with the expression of cyclinD2 and IGFBP2 minicking HL and relatively smaller tumor cells not expressing cyclinD2 and IGFBP2 imatating ALCL.Conclusions:1. Our study reported the different expression of cyclinD2 and IGFBP2 in protein level in HL and ALCL. High expression of cyclin D2 and IGFBP2 were mainly shown in HL but was low or negative expressed in most cases of ALCL. The expressing feature of cyclinD2 and IGFBP2 might imply a different mechanism of oncogenesis and growth control in HL and ALCL.2. In a few cases, pathohistological features, combined expressions of some lymphocytic markers, cyclinD2 protein and IGFBP2 protein in neoplastic cells showed an overlap and a likely transitional state between HL and ALCL.3. Prominent difference of the expression manner of cyclin D2 and IGFBP2 could be useful to the differential diagnosis between HL and ALCL. |
