Cordyceps sinensis is the dry complex of stroma of Cordyceps sinensis and the body of hepialidae larva. It has the effect of reinforcing kidney nourishing lung, hemostasis and resolve phlegm, for clinical kidney deficiency, impotence, nocturnal emission, waist and knee pain, cough and asthma, labor cough, hemoptysis, which is a traditional precious Chinese medicine. In this paper, different batches of Cordyceps sinensis and Cordyceps main adulterants is for the study. Establishing Cordyceps quality control measures includ a water-soluble ingredients fingerprints and determination of nucleosides. Identified the compounds that reprented by common peaks and characteristic peaks in fingerprints. Analyzed and Identified the main chemical constituents of Cordyceps sinensis. This studies provide a method and basis for Cordyceps sinensis quality control.In accordance with the relevant provisions, "Fingerprints of TCM injections technical requirements", Cordyceps sinensis and its major adulterants water-soluble ingredients fingerprint was established. By examining the analytical method, fingerprint analytical condition was established:GL WondaSil C-18 (250mm 4.6mm, 5 m) column was use, mobile phase was acetonitrile and 0.1%aqueous phosphoric acid gradient elution, detection wavelength was 260 nm, the flow rate was 0.8 mL/min, the injection volume was 20 μL, the column temperature was 25℃, the detection time was 60 min. The methodological study proves this method was accurate and reliable. Fingerprint pattern diagram was established, marking a total of 16 common peaks. By HPLC/MS techniques was used to identify one of the 12 peaks, respectively, cytosine, uracil, cytidine, hypoxanthine+ guanine, uridine, thymine, adenine, inosine, guanosine, thymidine, adenosine and cordycepin. Two characteristic peaks of Cordyceps sinensis T1 and T2 was identified, T1 was tryptophan component, T2 was a leucine or isoleucine trimer. T2 was marked and identified for the first time from Cordyceps sinensis fingerprint and has important significance of distinguishing and controlling quality of Cordyceps sinensis.11 kinds of nucleoside component content in Cordyceps sinensis and its adulterants were determined. Uridine, inosine, guanosine, adenosine is the highest content of four kinds of nucleoside component, accounting for more than 85% of the total ingredients nucleosides. Based on the differences in nucleoside component content of Cordyceps sinensis and main adulterants, a quality control standards of a uridine, inosine, guanosine, adenosine content as an indicator was established. That is the sample has to contain uridine, guanosine, inosine, and adenosine amount of not less than 0.28%, uridine contain not less than 0.12%.
