Cordyceps sinensis may provide ideas for the development of new drugs for the medical field, which has antibacterial activity with antibacterial active ingredients. Exploring antimicrobial composition and antibacterial properties of Cordyceps sinensis, in this study, we obtained fermentation products of it by liquid fermentation, after separation and purification of fermentation products were screened out components of antibacterial activity, and analyze comprehensive pathogens with antibacterial properties, finally, qualitative and quantitative analysis of the composition of antimicrobial ingredient and determination of the yield portion of the material under the test conditions by HPLC.Cordyceps sinensis be cultured by liquid fermentation to obtain fermentation products. Used by centrifugated, ultrasonicated, ethanol precipitated, deproteinized by Sevag, etc., We got fermentation broth, mycelium extracts, fermentation broth products though ethanol extract, soluble ingredients of fermentation broth, ethanol extracts of mycelium extracts, soluble ingredients of mycelium extracts, intracellular polysaccharides, extracellular polysaccharides.21strains of nonstandard pathogens and8strains of standard pathogens do antimicrobial tests. Testing the antimicrobial properties of the different components in fermentation products by filter paper diffusion method. The results showed that ethanol extracts in fermentation broth has a good broad-spectrum antimicrobial on bacterial pathogens, no significant inhibitory effect on yeast or fungal pathogens, and further explain showed that antibacterial property of the antimicrobial components combined with the antibacterial spectrum, possible development of new natural product drugs used for medical field.Preliminary analysis to composition of the antibacterial components and qualitative and quantitative analysis of some substance use by HPLC. The results showed that the antimicrobial components in the major nucleosides:cordycepin, hypoxanthine, a small amount of adenosine, and suspected substances:guanine, xanthine, cytidine. The yields of several substances in fermentation broth under the experimental conditions were:cordycepin2.85mg/100mL, hypoxanthine2.34mg/100mL, adenosine0.62mg/100mL, suspected guanine3.36mg/100mL, suspected xanthine0.64mg/100mL and suspected cytidine0.79 mg/100mL.In addition, ethanol extracts of mycelium extracts have no antibacterial activity with corresponding ethanol extracts in fermentation broth were analyzed by HPLC. The results showed that the main nucleosides of ethanol extracts of mycelium extracts were:adenosine, a small amount of adenine and cordycepin, suspected substances:uridine, thymine, xanthine and inosine. There have large differences in the composition of matter compared with ethanol extracts in fermentation broth. The yields of several substances in mycelium under the experimental conditions were:adenosine5.07mg/g, adenine0.28mg/g, cordycepin0.22mg/g, suspected uridine0.26mg/g, suspected thymine4.12mg/g, suspected xanthine0.25mg/g and suspected inosine3.74mg/g. |
