Objective:In view of preliminary,research, this experiment further probed themolecular mechanism of Tongmaidasheng-Tablet(TMDST) based onBuShen TiaoChong on regulating gonadotropin receptors in ovary and hemorheology of rats with ovulation failure caused by kidney deficiency,to evaluatetheimpact of BuShen TiaoChong on ovarian local environment,and provide theoretical basis andobjective evidence for the clinical application.Method:According to the modeling method of Jin Yu, the model of ovulation failure caused by kidney deficiency was established by injecting testosterone propionate subcutaneously to SD female rats of 9 days age. Then the rats were divided into seven groups:model group,TMDST high-dose group,TMDST medium-dose group,TMDST low-dose group,letrozole group,You Gui-pill group and blank control group. The medication was started at 80 days old until 5weeks later,then the blood was taken through arteryfemoralis to detect The content of hemorheology indexes and serum TGF-β1.Ovary was harvested,weighed up after dead.After froze by liquid nitrogen,the rats was used to detect the expression of Follicle-Stimulating Hormone receptor mRNA(FSHR mRNA)and Luteinizing HormonemRNA (LHR mRNA) by Real-time PCR.Results:1.The weight change of model group were lower than blank control group(.P< 0.05). The weight change of TMDST high-dose and letrozole group were higher obviously (P< 0.01);that of TMDST medium-dose group and You Gui-pill group were also higher(P <0.01),in comparison with that of model group.Among different dose of TMDSTgroup,The weight change of high-dose group were highest.That of Letrozole group were gigher than that of blank control group(P<0.01).2.The overyindex ofmodel group was lower than blank control group(P<0.01).That of TMDST high-dose group,medium-dose group,You Gui-pill group and letrozole group were all higher than that of model group(P< 0.01). TMDST low-dose group had no significant change. Among all group of drugs,the overyindex ofletrozole group was highest.3. The FSHR mRNA’s expression ofmodel group was lower than blank control group(P< 0.01). The FSHR mRNA’s expressionof TMDST high-dose group and letrozole group were higher obviously than that of model group(P< 0.01); that of TMDST medium-dose group,low-dose group and You Gui-pill group also rised(P< 0.05).4. The LHR mRNA’s expression level of model group was lower than blank control group(P< 0.01).The LHR mRNA’s expression level in groups of TMDST high-dose, medium-dose,letrozole and You Gui-pill were higher than that of model group(P< 0.01);that of TMDSTlow-dose group also rised(P< 0.05).5. The level of TGF-β1 in model group.model group was lower than blank control group(P<0.01). That of letrozole group rised obviously(P< 0.01);that in groups of TMDST high-doseandand You Gui-pill were higher(P< 0.05);that in groups of TMDST medium-dose and low-dose had no significant change(P>0.05),in comparison with that of model group. Among different dose of TMDSTgroup,the level of TGF-βl in high-dose group was highest.6.There was positive correction not only betweenthe expression of FSHR mRNA and The level of serum TGF-β1 (r=0.721, P<0.01),but also the expression of LHR mRNA and The level of serum TGF-β1(r=0.876, P<0.01).7.①In themodel group,the blood viscosity at low shearrate and that of high shearrate elevated (P< 0.05);red cell deformability(RCD) index declined(P< 0.05);other indexes had significant change(P>0.05).②In comparison with model group,the blood viscosity at low shearrate of TMDST high-dose group declined significantly(P<0.01); the blood viscosity athigh and median shearrate,plasma viscosity and RCD index of TMDST high-dose group changed positively(P<0.05).the blood viscosity and plasma viscosity of TMDST medium-dose group declined significantly(P< 0.05).The hemorheology indexes of TMDST low-dose group,letrozole group,You Gui-pill group and blank control group had no significant change(P>0.05).Conclusion:the mechanism of Tongmaidasheng-Tablet(TMDST) improving follicular developmentand ovulation based onBuShen TiaoChong was achieved through multi-link and multi-target.1. BuShen TiaoChong could improve dysgenic and dysplasia by perfecting the weight change rate and overyindex in favour of follicular development.2.There was imbalance Gn receptor (FSHR,LHR) mRNA of ASR.TMDST could upregulate the expression of Gn receptor (FSHR,LHR) mRNA,to improve reaction of ovary to FSH and LH,improving follicular developmentand ovulation.Effect of TMDST high-dose group was best.3. the influence mechanism of TMDST on the expression of Gn receptor mRNA was probably related to regulation on the expression of TGF-β1.4. There was abnormal change in hemorheology of ASR.TMDST could regulate the abnormal condition of hemorheology,to optimize local microcirculationand ovarian function.
