Royal jelly is the food resource of bee queen during her lifetime and workers during first 3 days of larval stage. Bee pollen and honey were workers’ food resource in other time. Then, the lifespan of queen is more than 30 times longer than that of worker. So it is considered that royal jelly has anti-aging effects. The anti-aging effects of royal jelly were researched in this thesis. In this study,450mg/mL D-galactose was used 0.3mg/g body weight to enhance mice aging. The aging indicators of SOD、MDA、GSH-PX、NO of serum and tissue, the weight of the body, the weight of liver and spleen were determined to evaluate anti-aging effects of royal jelly, royal jelly protein and alcohol extract. The main contents and results of the experiments were as follows:1. Establishment of aging model mouse:the concentration of D-galactose injected in this study is 450mg/mL. D-galactose solution 0.3mg/(g*bw) of mice was injected cervical for four weeks. The SOD and MDA of serum were determined. The results were as follow: compared with the normal mouse, the SOD of serum and body weight significantly was reduced and the MDA of the serum was increased, which means that the aging model is successful.2. The anti-aging effect of Royal jelly:the experiment mice were divided into 13 groups,20 each group. They were normal control group, model control group and bovine serum albumin control group, royal jelly groups of different concentrations (1.05g/kg,2.10g/kg,3.15g/kg,4.20g/kg and 5.25g/kg). The normal and bovine serum albumin control group mice were separately fed with distilled water and 1g/kg bovine serum albumin. In each group,10 mice are anesthesia with anhydrous ether. Blood was collected from eyeballs of mice. Liver and spleen were separated and made as homogenate with 0.9% NaCl. The contents of SOD, MDA, GSH-PX and NO of serum and tissue, and weights of body, liver and spleen of mice were measured. Another 10 mice of each group were used for the hypoxia tolerance test. The results were that: royal jelly significantly increased the level of SOD and GSH-PX of the serum and tissue, body weight and weight of the spleen of aging mice, reduce the level of MDA, NO and the weight of liver, extended hypoxia tolerance of aging mice and the resistance ability of mice.3. The anti-aging effect of royal jelly protein and alcohol extract: Experimental mice were divided into 13 groups. There were 5 royal jelly protein groups,5 alcohol extract groups, normal control group, model control group and bovine serum albumin control group. The doses of royal jelly protein were 0.12g/kg,0.24g/kg,0.36g/kg, 0.48g/kg and 0.60g/kg. As to alcohol extract those were 0.018g/kg,0.036g/kg, 0.054g/kg,0.072g/kg and 0.090g/kg. The contents of SOD, MDA, GSH-PX and NO of serum and tissue and the body weight, liver weight and spleen weight of mice were tested after four weeks continuously feeding. The results were that:Royal jelly protein and alcohol extract increased the body weight and the weight of the spleen of aging mouse, but reduced the weight of liver of aging mice. Royal jelly protein significantly increased the level of SOD and GSH-PX of the serum and tissue of aging mice, reduced the levels of MDA and NO of the serum and tissue, and prolonged hypoxia tolerance of aging mice and stress resistance ability of mice. Alcohol extract increased GSH-PX of the serum and tissue of aging mice. From the test, it can be deduced that royal jelly protein could prolong the lifespan of the aging animal in certain degree.
