Objective:To study the protective effect of apigenin in myocardium of rats with isehemia reperfusion injury.To further clarify whether the protective effect of apigenin against anoxia/reoxygenation injury mediated by Bcl-2.Method:In this study, the A/R modle was replicated on H9c2 cardiomyocyte by anoxia/reoxygenation treatment. H9c2 cardiomyocyte randomly divided into four groups, each group repeated six times:(1)Control group; (2) A/R group; (3) Apigenin groups:Api-10+A/R,Api-20+A/R,Api-40+A/R,Api-80+A/R,Api-160+A/R group; (4) Api-40+ABT-737+A/R group.The cell viability was detected by MTT; Western blotting analysis assayed the expression of Bcl-2; The DCFH-DA assayed the level of intracellular reactive oxygen species ROS; According to the kit,the activity of SOD,LDH,and GSH-Px was analyzed with an automatic biochemical analyzer; The content of MDA was analyzed by the TBA; the mitochondria membrane potential were measured by the JC-1; the percentage of apoptosis were measured by annexin V-FITC/PI double staining.Results:1.In different dose apigenin groups compared to A/R group, the Api-40 +A/R group on cell viability was significantly increased.2. After Api-40+A/R, the expression of Bcl-2 protein after processing A/R was increased compared to A/R group.3. After Api-40+A/R, the cell viability and the activity of SOD and GSH-Px were increased while the activity of LDH, the generation of ROS, the content of MDA,the mitochondria membrane potential and the apoptosis were decreased. However, in the Api-40+ABT-737+A/R group, the cell viability and the activity of SOD and GSH-Px were decreased while the activity of LDH, the generation of ROS, the content of MDA,the mitochondria membrane potential and the apoptosis were remarkably increased compared with A/R groupConclusion:Cardiomyocytes protecton of apigenin against anoxia/reoxygen-ation injury mediated by Bcl-2.
