Objective:Apigenin is the widely distributed flavonoid that exits in vegetable,traditional chinese medicinal materials. In recent years,considerable attention has been devoted to research its pharmacological effects. Some researchers have found that apigenin has protective effects against myocardial ischemia reperfusion injury. The mechanisms of apigenin to protect cardiomyocytes from ischemia reperfusion injury may be assossiated with intracellular signal pathways and the regulation of proteins expression. But the mechanism underlying this cardioprotection remains largely unknown. In the study,we established myocardial A/R injury model and the adenovirus efp- si RNA to deeply explore the relationship between the effect of apigenin against myocardial hypoxia/reoxygenation(A/R) damage and the efp protein.Methods:The cultured H9c2 myocardial cells were divided into 5 groups randomly and each group was repeated six times:(1) Control group;(2) A/R group;(3) apigenin +A/R group;(4) apigenin + negative control +A/R group;(5) apigenin + efp-si RNA +A/R group. The expression of efp was assayed by western blotting analysis and the cell viability was detected by MTT. The flow cytometry was used to detect the degree of cell apoptosis,the intracellular ROS and the mitochondria membrane potential.Used the UV spectrophotometer to analyze the content of Methane Dicarboxylic Aldehyde(MDA) and the activities of superoxide dismutase(SOD), lactate dehydrogenase(LDH)and glutathione peroxidase(GSH-Px).Results:Apigenin pretreatment group increased the level of efp protein compared with the control or A/R group(p<0.01). While apigenin + efp-si RNA + A/R group didn’t increase the expression of efp; moreover,apigenin pretreatment group could increase the viability of cardiomyocytes treated with A/R, decrease the generation of reactive oxygen species(ROS),maintaine the mitochondrial membrane potential(ΔΨm) andattenuate cell apoptosis induced by A/R injury.however these protective effects partly completely were abrogated in the group pretreated with apigenin + efp- si RNA + A/R group.Conclusion:The effect of apigenin against cardiomyocytes anoxia/reoxygenation injury depend on(at least partly depend on) regulating the expression of efp protein.
