Experimental Study Of Effect Of Rhunbarb Zhechong Pill On The Immune Status Of Tumor-bearing Mice With Lewis Lung Carcinoma Intervention

Objective:According to the theory of Traditional Chinese Medicine that "the evil of the hash, the gas will empty"and "cancer what is also rock",lung cancer is caused by blood stasis. So we select the Rhunbarb Zhechong Pill which can support the rational existence of deficiency and stasis of lung cancer as an intervention for lung cancer. We can study the treatment and immunomodulatory effects of Rhubarb Zhechong pills on Lewis lung cancer tumor-bearing mice and provide experimental evidence for the clinical treatment of cancer.Methods:64 SPF Kunming mice, half male and half female, were divided into the normal group, the model group, the Rhunbarb Zhechong group(Traditional Chinese Medicine group), and the Cisplatin group (Western medicine group) randomly. The normal group didn’t take any measures, while the mice of the other three groups were subcutaneous injected with 0.2ml of Lewis Lung Cancer (LLC) cells suspension in their right armpit. Drug were intervened after three days after modeling. Then all the mice were weighted sacrificed at 29th day. Taking the whole lung, making it into slice and hematoxylin-eosin staining and observing it in the optical microscope. Weighing tumor and calculating the inhibition rate. Thymus and spleen were weighed and thymus index and spleen index were calculated. The levels of serum interleukin 2(IL-2) were measured by Enzyme-linked immunosorbent assay (ELISA) and the ratio of CD4+CD25+Foxp3+were detected by Flow Cytometry.Results:1 Morphological changes:Observing the lung slice of each group under the optical microscope, the normal group, the Traditional Chinese Medicine group (TCM group) and the Western medicine group were found alien cells in the lung. And there is no alien cells in the mice of the normal group.2 Weight change:Compared with the normal group, the weight of the other three groups significantly decreased(P<0.01). Compared with the model group, the weight of the TCM group and the Western medicine group exist differences significant (P<0.05). The weight of the TCM group was significantly higher than the model group’s. And theweight of the Western medicine group was significantly lower than the model group’s. The weight growth trend of each group of mice is the normal group>the TCM group>the model group>the Western medicine group.3 Organ index:Compared with the normal group, the Thymus index in the model group, TCM group, Western medicine group significantly decreased (P<0.01). Compared with the model group, the Thymus index in the TCM group significantly increased (P< 0.05), and it decreased in the Western medicine group (P<0.05).Compared with the normal group, the Spleen index in the model group, the TCM group increased (P<0.05) and decreased in the Western medicine group (P<0.05). Compared with the model group, the Spleen index in the TCM group decreased (P<0.05).4 Inhibition rate:Compared with the model group, the tumor weight(P<0.05) in the TCM group, Western medicine group significantly reduced.5 The ratio of CD4+CD25+Foxp3+in spleen:Compared with the normal group, the ratio of CD4+CD25+Foxp3+in spleen in the other three groups significantly increased (P< 0.01). Compared with the model group, the ratio of CD4+CD25+Foxp3+ in two treatment groups significantly decreased (P<0.05).6 The levels of serum interleukin 2 (IL-2):Compared with the normal group,the levels of serum interleukin 2 (IL-2) in the other three groups significantly decreased (P< 0.01).Compared with the model group the levels of serum interleukin 2 (IL-2) significantly in the TCM group increased (P<0.05) and decreased in the Western medicine group (P< 0.05).Conclusion:1 Injection of Lewis lung cancer cells in mice can lead to the formation of tumor-bearing mice.2 Rhubarb Zhechong pill can suppress the lung tumor growth of the mice injected Lewis lung carcinoma. And Rhubarb Zhechong pill can protect the body’s immune organ.3 Rhubarb Zhechong pills suppress the lung tumor growth by reducing the proportion of CD4+CD25+Foxp3+/CD4+4 Rhubarb Zhechong pill have immunomodulatory effects on the mice with Lewis lung. It can increase IL-2 levels and enhance immune function of the tumor-bearing mice with Lewis lung carcinoma cells.
