Gastric diseases have been a problem perplexing people. Reseaches have shown thatH.pylori is the major reason. The infection of H.pylori can cause gastritis, and the severecases may develop to gastric ulcer and gastric cancer. At present, the effect of eradicationtherapy of H.pylori is poor; the development and progression of H.pylori-infected gastridiseases is still not clear. Therefore, improvement of the immunological effect hasbecome the on of the hotspots in the reseach field. CD4+CD25+Foxp3+regulatory T cell,playing a negative role on the development, progression and the prognosis of gastricdiseases, has two powerful functions: immune anegy and immunosuppression. Reseacheshave confirmed melatonin (MLT) contributes to anti-tumor activity and immuneregulation. It is confirmed MLT could reduce the weight and volum of tumor, decreasethe number of Tregs and its transcription factor Foxp3expression in the tumor tissue.MLT also could down-regulate the number of Tregs and scurfin in the thymus and spleen,and up-regulate Foxp3mRNA expression in spleen. The data indicates one of MLTanti-tumor way is via inhibition of Tregs in vivo[1].In this reseach, the main object is toexplore the influence of MLT on gastric diseases infected by H.pylotic and therelationship between MLT and Treg cells. Thus, we try to find a new idea based onNeuro-Endocrine-Immune network for H.pylori-infected gastric diseases; and offerrelative clues for therapy of diseases by MLT.The study establishes the mice model bearing gastric diseases infected by H.pylori.After the intervention of MLT at different concentrations, we detect the expression ofCD4+CD25+Foxp3+Treg cells and Foxp3mRNA in thymus and spleen by FCM andReal-time PCR at2w、4w and6w. We detect the pathological injury level of stomach byHE staining, the invation level of H.pylori by rapid urease test after culturing H.pylorifrom gastric tissuse, and verify the colonization of H.pylori by his red dye.The resultsshow the gastric mucous and submucosal layer have benn suffused with inflammatory cells obviously. H.pylori chould be seen tip sample colonies on the blood plate, and itsmorphology in microscope by his red dye.MLT could down-regulate the expression oftranscription factor Foxp3mRNA in thymus, and the number of Tregs and itstranscription factor Foxp3expression in spleen. The data indicated H.pylori hassuccessfully infected the stomach of mice; and on of the way of MLT influence on gastricdiseases is to decrease the number of Tregs.In conclusion, MLT could have influence on the severity of H.pylori-infected gastricdiseases, and the mechanism maybe concerned with the down-regulation of number ofTregs and expression of transcription factor Foxp3mRNA.
