Objective:To analyze the role and clinical significance of HIF-2α、 CITED2 and PTPRZ1 during the development of papillary thyroid carcinoma (PTC) by detecting the expressions of HIF-2α、CITED2 and PTPRZ1 in the PTC tissues;The study aims to explore the mechanism of action which HIF-2a plays a role in the PTC and also to search the diagostic marker of PTC and provide the new idea to the diagnosis and treatment of thyroid carcinoma.Methods:1 Cases collection and grouping:50 cases of PTC specimens and blood (PTC group),10 cases of thyroid adenoma specimens and blood(TA group),60 cases of normal tissues adjacent to the tumors and 10 cases of healthy persons (N group) with integrity clinical pathological datas were collected from April 1,2014 to April 1,2015 year2 The expressions of HIF-2α、 CITED2 and PTPRZ1 proteins in PTC, TA and normal tissues adjacent to the tumors were detected by Western blot and immunohistochemical SP method.And the relationships with the clinical pathology characteristics were analyzed combined with the integrity clinical pathological datas. The transcriptional level changes of HIF-2amRNA、CITED2mRNA and PTPRZ1mRNA were tested by real-time reverse transcription polymerase chain reaction.3 All the statistical analyses were carried out using SPSS17.0. The datum were expressed as x±s and analyzed by single factor analysis. The positive rates of immunohistochemistry were compared by chi square test. Bilateral P<0.05 was considered statistically significance.Protein expression of Western Blot was got by using Imaging J to detect region and gray value of protein expressionResults:1 The results of immunohistochemistry staining showed HIF-2α、CITED2 and PTPRZ1 proteins were expressed in different strength in all PTC tissues. Their positive proteins were mainly located in the cancer cell cytoplasm, a few or not were found in the normal tissues adjacent to the tumors or thyroid adenoma tissues. Their positive expression rate in PTC tissues are obviously higher than that in TA group and N group(P<0.01) and expressions between N and TA were no significant difference (P>0.05). The expressions of HIF-2α、CITED2 and PTPRZl were unrelated to the age, sex and size of the tumor (P>0.05) of PTC patients, but associated with the lymph node metastasis and pathological stage (P<0.05).2 The results of Western Blot showed the HIF-2α、CITED2 and PTPRZ1 proteins in PTC tissues were significantly more than in TA and normal tissues adjacent to the tumors. (P<0.05), but there was no statistical difference between in TA and normal tissues adjacent to the tumors (p> 0.05).3 The results of Real-time PCR showed the transcriptional expression levels of the HIF-2a mRNA、CITED2 mRNA and PTPRZ1 mRNA in PTC tissues were significantly higher than in TA and normal tissues adjacent to the tumors with statistical significance(P<0.05), but there was no statistical difference between in TA and normal tissues adjacent to the tumors (P>0.05).Conclusion:HIF-2α、CITED2 and PTPRZ1 are all detected in PTC TA and normal tissues adjacent to the tumors,but expressons are stronger in PTC tissues than in TA and normal tissues adjacent to the tumors.The higher expressions of HIF-2α、CITED2 and PTPRZ1 in PTC have the correlation with the lymph node metastasis and clinicopathologic stage, HIF-2 a may promote the occurrence, development, invasion and metastasis of PTC by the higher expression of CITED2 and PTPRZ1.All of them play the important role in the process of PTC, include the occurrence, development,invision and metastasis.
