Objective: To explore the relationship between polymorphism loci of vascular endothelial growth factor(VEGF)-2578C/A gene and breast cancer and to analyze the correlation between polymorphism loci of VEGF-2578C/A and the body’s susceptibility to breast cancer from the gene level, in order to clarify the pathogenesis of breast cancer in molecular biological level and assist the study on the pathogenesis of breast cancer.Methods: Breast cancer patients(confirmed by postoperative pathological results) were selected from those admitted in Chengde Center Hospital between October 2011 and December 2013 to form the case group, aging from42 to 75 years old, 54.18±9.08 years on average. Healthy people who had physical examination in the physical examination center of Chengde Center Hospital were selected as the control group, aging from 31 to 78 years old,53.30±9.31 years on average. Fasting peripheral venous blood was taken on the morning to extract DNA. Ligase detection reaction(LDR) was used to detect the genotype of VEGF-2578C/A and SPSS 19.0 was used for chisquare test on the data and the assessment on the relative risk of the disease.P<0.05 referred to statistical significance of the results. The two groups were compared for the frequency of genotype, allele frequency, the odds ratio(OR)and 95% confidence interval(95% CI) to analyze the correlation between polymorphism loci of VEGF-2578C/A gene and breast cancer and to discuss the correlation between this polymorphism loci and the body’s susceptibility to breast cancer.Results:1 Frequencies of the three genotypes(CC CA AA) of VEGF-2578C/A gene polymorphism loci were 83.75%, 12.5% and 3.75% in the breast cancer group, while the corresponding frequencies in the healthy control group were50.0%, 50.0% and 0.00%. There were statistically significant differences between the two groups(P<0.01) in distribution of genotype frequency; and compared with genotype CC, genotype CA could increase the susceptibility to breast cancer.2 Distributions of allele frequencies(C, A) were(90.00%, 10.00%) and(75.00%, 25.00%) in the breast cancer group and the healthy control group.There were statistically significant differences between the two groups(P<0.01, OR>1); and there were significant differences between allele C and allele A(X2=9.412, P=0.002, OR=3.000, 95% CI of 1.456-6.182). And allele C could increase the risk of breast cancer.Conclusion:1 There may be a certain correlation between the polymorphism loci of VEGF-2578C/A gene and the risk of breast cancer.2 VEGF-2578 CA genotype could increase the susceptibility to breast cancer.3 VEGF-2578 C allele can increase the risk of breast cancer.
