MiRNAs are a class of small endogenous non-coding RNAs, 22-23bp in length, which bind with the 3’-UTR of targets mRNA and induce the silencing complex to degrade the target mRNA or inhibit the translation of the target mRNA. miRNAs play an important role in organ development, cell proliferation, apoptosis, tumorgenesis, metastasis and many other life activities. miR-17-92 is a highly conversed and polycistronic miRNA cluster, which encodes for 6 individual miRNAs. miR-17-92 cluster regulates many genes which play an important role in biological development, so the research of miR-17-92 cluster in depth has very important significance. Semaphorin 5A is involved in the regulation of axonal guide, cell proliferation, apoptosis, cell migration and other physiological functions though combinate with PLXNB3, the receptor of SEMA5A.In our system, we predicted that SEMA5A is a potential target gene of miR-18a, referring to the bioinformatics analysis. And luciferase reporter assay confirmed the interactions. Real-time quantitative PCR and Western blot proved that SEMA5A expression was reduced at mRNA and protein levels when miR-18a was overexpressed in the gastric cancer MKN45 cells. Meanwhile, we found that overexpression the miR-18a inhibits the process of cell proliferation and migration. Overexpression the miR-18a and SEMA5A could rescue the process of cell proliferation and migration through the wound healing assay and CCK-8 assay, which indicated that miR-18a inhibits the process of cell proliferation and migration mediated the regulation the SEMA5A. Moreover, we excluded the probable effect of on MKN45 apoptosis by flow cytometry.
