Aim: microRNA moledule plays an important regulatory in the immune response between infection of the pathogen and anti-infection of the organism.Previous studies have showed that some miRNAs were significantly in body fluid and cell of patients with tuberculosis.To explore the expression levels and the significance of microRNA-125b(miR-125b),microRNA-29a(miR-29a) and microRNA-155-5p(miR-155-5p)in cerebrospinal fluid(CSF)and plasma from the patients with tuberculous meningitis.Methods:All 40 cases of patients were selected from Shihezi Medical University Affiliated Hospital,Shihezi People ‘s Hospital and Yili Kazak Autonomous Prefecture of Kuitun Hospital during October 2013 to June 2014.Twenty cases of the clinical diagnosed tuberculous meningitis patients called tuberculous meningitis group. The twenty tuberculous meningitis patients,15 are male,5 are female,the average age is 42.95±16.33 years.There are2 spinal tuberculosis and 2 cavitary pulmonary tuberculosis.The diagnosis criteria is according to clinical diagnosis criteria suggested by Thwaites.Twenty cases of surgery patients without neurological disorders called control group.The twenty control patients,11 are male,9 are female,the average age is 44.50±19.40 years.Cerebrospinal fluid and plasma samples were collected from tuberculous meningitis group and control group,and the total RNA was extracted respectively.The levels of miR-125 b,miR-29 a and miR-155-5p were measured by real-time quantitative PCR(qRT-PCR).Results:The results of qRT-PCR showed the existence of differences in expression of miR-125 b,mi R-29 a and miR-155-5p in peripheral blood plasma of tuberculous meningitis group and control group.meningitis group significantly higher than control group, which had statistical significance(P<0.01); ⑵ the level of miR-125 b in both CSF and plasma of tuberculous meningitis group significantly higher than control group, which had statistical significance(P<0.01).Conclusion:(1)The study found that the expression of miR-125 b,miR-29 a and miR-155-5p was very differential in plasma in tuberculous meningitis group,and speculated that miR-125 b,miR-29 a and miR-155-5p may participate in the immune reaction of tuberculous meningitis.(2)The levels of miR-29 a and miR-125 b in both CSF and plasma of tuberculous meningitis group significantly higher than control group. It suggested that miR-125 b and miR-29 a may be as potential biomarkers for detecting tuberculous meningitis.
