Objective: To investigate and compare the ptotein expression level of serum fos-like antigen2(Fosl 2)gene and correlated analysis with clinical features of type 2 diabetes mellitus(T2DM)in Uyghurs in Xinjiang, and provide a basis for illustrating the pathogenesis and developing the treatment plan of T2 DM in Xinjiang ethnic minorities.Methods: A total of 50 Uyghurs patients that diagnosed with T2DM(T2DM gruop) and 50non-diabetes mellitus Uyghurs(NDM gruop)were collected from Kashi region’s hospital in the study. The data of sex, age, weight, height, waist, hip, blood pressure and course of T2 DM were measured and record,then calculate the body mess index(BMI) and waist hip radio(WHR).The fasting plasma glucose(FPG) and lipis were simultaneously detected by automatic chemistry analyzer.The level of fasting insulin(FINS) was tested by chemiluminescent immunoassay.The level of glycosylated hemoglobin(Hb A1c)was measured by high pressure liquid chromatography(HPLC). To get the insulin sensitive index(ISI),HOMA-IR and HOMA-β by using the fomula. The protein expression of Fosl 2 gene were surveyed by enzyme-linked immunosorbent assay(ELISA).The data were analyzed by SPSS19.0 statistical software and represented through mean ± standard( X ± S) deviation,meanwhile the data were tested normality and homogeneity of variance. If the data were showed by a skewed distribution, it should be taken the natural logarithm before doing the statistical analysis.Independent-samples t test were used between the two groups.The relationships between the protein expression of Fosl 2 gene with other indicators were analyzed by pearson correlation analysis and multiple linear stepwise regression analysis.Results:1. Comparing with Uyghurs NDM group,the clinical basic data, such as age, sex radio,DBP, SBP, WHR had no statistically significant differences in Uyghurs T2 DM group(P>0.05).2. Comparing the blood and lipids indicators with Uyghurs NDM group, the levels of FPG(5.45±0.69,11.46±4.40;P<0.001), Hb A1c(6.21±1.30, 10.20±2.10;P<0.001),T C(3.80±0.87, 5.16±0.87;P<0.001),TG(1.48±0.85, 2.21±1.02;P<0.001), LDL-C(1.48±0.85, 2.21±1.02;P<0.001)were significantly higher in Uyghurs T2 DM group,while the HDL-C was lower(1.20±0.30, 1.03±0.34;P<0.05); in the comparison of the inslin resistance indexs between Uyghurs NDM and T2 DM group, the FINS( 11.61±6.67,16.13±11.41;P<0.05)and HOMA-IR(2.74±1.59, 7.18±5.32;P<0.001)were higher in T2 DM group;in the comparision of islet beta cell function, the ln ISI(-3.9±0.80,-4.93±0.73;P<0.001)and ln HOMA-β(4.63±0.85, 3.63±0.96;P<0.001)were obviously lower.3. Comparing with Uyghurs NDM group, the protein expression of Fosl 2 gene was apparent lower in T2 DM gruop(41.48±26.32,18.09±9.48;P<0.05).4. The correlation analysis results showd,in Uyghurs NDM control group, the protein expression level of Fosl 2 gene had no correlation with age,waist, height, hip, weight,WHR, BMI, FPG, TC, TG, HDL-C, LDL-C, FINS, HOMA-IR and other indicators(P>0.05);however,in Uyghurs T2 DM group, the serum protein expression level of Fosl2 gene was positively correlated with DBP(r=0.445,P<0.05)and TG(r = 0.536,P<0.05),and negatively correlated with HOMA-β(r =-0.469,P<0.05)and FINS(r =-0.465,P<0.05).5. In multiple stepwise linear regression analysis, the ptotein expression of Fosl 2 gene level as dependent variables, the levels of age, weight, height,WHR, BMI, SBP, DBP, FPG, TC,TG, HOMA-β and other indicators as independent variables,the results displayed DBP and BMI went into the regression equation,that reminder they may be the main factors of influencing the protein expression of Fosl 2 gene.Conclusions:1. In Xinjiang Uyghurs population, comparing with NDM group, the T2 DM patients had clearly glucose and lipis disorder.2. Comparing with Uyghurs NDM group, the protein expression level of Fosl 2 gene was reduced in the T2 DM patients, that means the Fosl 2 gene may involve in the process of Uyghur T2 DM, moreover, the DBP and BMI may be the factors that affect the protein expression of Fosl 2 gene of Uyghurs with T2 DM in Xinjiang.3. The Fosl 2 gene expression level has relation with insulin resistance(IR) and islet β cell function in Uyghurs T2 DM population, moreover, it could be lay the foundation as a indicator to evaluate IR and β-cell function of Uyghurs T2 DM in Xinjiang in future.
