Abstract/Summary:
Objective: To observe the expression characteristics of epidermal ERAP1 protein in psoriasis vulgaris, a preliminary verification of whether ERAP1 is involved in the progress of pathogenesis of psoriasis vulgaris,for the further study and treatment of psoriasis and provide the theoretical basis and experimental basis.Methods: The clinical pathological diagnosis in our department for "consistent with psoriasis" and according to the clinical stage of standard classification of typical progress and stability of typical psoriasis vulgaris paraffin tissue specimens a total of 49 cases, advanced stage 23 cases, stable stage 26 cases,ovarian cancer tissue as positive control, PBS as negative control and 3 cases of normal skin tissues, to detect expression of ERAP1 protein by immunohistochemical method, progress and stability are expressed and compare the expression difference. SPSS13.0 Statistical analysis was performed on epidermal ERAP1 protein in psoriasis vulgaris lesions.Results: In addition to the typical histopathological signs(thickening of the epidermis, epidermal sudden extension, hyperkeratosis with incomplete, horny cell nuclear mitotic activity increased infiltration of lymphocytes, etc.), advanced stage, stable stage of psoriasis vulgaris tissue were seen in epidermal ERAP1 expression in the cytoplasm, brown particles, but the expression characteristics of the existence of differences by the study of characteristics, divided into the Department of Dermatology, senior doctors assess the expression of ERAP1 protein in the cytoplasm of epidermal cells within the epidermis tissue: 1diffuse positive expression of type, 2 upper, lower epidermis of positive expression of strong weak type.Compared with the advanced stage, stable stage of ERAP1 protein was mainly in epidermis of diffuse,uniform expression, the coincidence rate was 84.6%(22/26); compared with the period of stability,progress of phase distribution in the epidermis does not diffuse uniformity, and to the upper, lower epidermis strong weak expression, the coincidence rate was 82.6%(19/23). The use of SPSS13.0 software of the data were corrected for 2×2 contingency table chi square test, the results had significant difference(χ2=19.53, P=0.000, OR=26.16).Conclusion: 1. ERAP1 expression in different psoriasis vulgaris stages, and feature difference in preliminary indicated that the protein and GWASs screening susceptibility genes for psoriasis consistency,and is related with the stage disease;2. ERAP1 in typical period and stable period psoriasis expression are differences, and the former showed a trend of lower epidermis than the latter to express different level drops or missing.Ovarian cancer lymph node metastases from primary lung cancer site ERAP1 express decline or missing, explain ERAP1 expression characteristics are associated with the deterioration of the disease at different stages and ease;3. Based on the ERAP1-peptide-MHCⅠ-CD8+T cells relations, ERAP1 expression differences show the former than the latter in lower epidermis to express different level decline or loss may be related to the channel such as CD4+ and CD8+T other molecules mediated disease occurrence and development together.
