Objective To investigate the effects of astragaloside IV on neointima hyperplasia in the carotid artery of rats after ballon injury and explore the inhibitory mechanisms. Methods Fifty healthy male Sprague-Dawley rats were randomly divided into five groups:a sham-operation group(sham), a model group(model), and three astragaloside Ⅳ-treated groups. Three days before the surgery,1% CMC, astragaloside IV 20,40, 60 mg·kg-1 were administered by intragastrically in sham, model and 3 astragaloside-treated groups once daily for 17 days. Fourteen days after the surgery, rats were killed, and the carotid arterys were harvested. Hematoxylin-elsin staining was carried out to observe the pathomorphological change, and presented as the neointimal and media area ratio. Immunohistochemistry staining was performed to measure the expression of proliferating cell nuclear antigen (PCNA), basic fibroblst growth factor (b-FGF), transforming groth factor-β1(TGF-β1), platelet-derived growth factor-BB (PDGF-BB. PCNA, b-FGF, TGF-β1 and PDGF-BB were analyzed with Iamage-Pro Plus. Plasma superoxide dismutase(SOD) activities and maleic dialdehyde(MDA) levels were detected. The SPSS 13.0 for windows statistical software was used for statistical analysis, All data are expressed as means±S.D. Only one-way analysis of variance was used to analyze the differences between two groups, P<0.05 was considered statistically significant.Results Compared with the model group, neointimal area, media area, and the expression of PCNA, b-FGF, TGF-β1 and PDGF-BB devreased significantly(p<0.05), the plasma SOD activity increased remarkably(p<0.05), while the MDA level reduced significantly(p<0.05) in astragaloside IV groups.Conclusion Astragaloside Ⅳ significantly inhibited neointimal hyperplasia in rat carotid artery 14 days after injury. It might be partially attributed to the inhibition of vascular smooth muscle mirgiation and proliferation, and the anti-oxidative effect.
