Abstract/Summary:
Objective:Observe the distribution and expression of KCC1 and b FGF in lens epithelial cells of different types of age-related cataract patients. Explore the roles and signaling pathways of K-Cl cotransporters in lens epithelial cellsMethods:1. We collected the anterior capsules which were in the age-related cataract lens, including 46patients(52 eyes) with age-related cataract. They were divided into three groups, in accordance with the location of turbidity in lens. The research object were the lens epithelial cells on the anterior capsules, a part of them were fixed by 4% paraformaldehyde and the others were cryopreserved and storage in refrigerator.2. To observe the distribution and the express trend of KCC1 and b FGF through Immunofluorescence assay.3. To test the two proteins expression through calculating the average relative gray-values of Western Blot.Results:1. Immunofluorescence assay: KCC1 and b FGF could both be finded in the cytoplasm of lens epithelial cells in different types of age-related cataract. The fluorescence intensity in lens epithelial cells of nuclear cataract group was stronger than the other groups; this showed that the expression level of KCC1 and b FGF in nuclear cataract group was highest.2.Western blot: The relative expression quantity of nuclear cataract group was larger than that of the other two groups,and the difference between nuclear cataract group and cortical cataract group was statistically significant(P<0.05). Strong and positive correlation was found among b FGF and KCC1,(P<0.01, r=0.875).Conclusion:1. KCC1 and b FGF were both expressed in the cytoplasm of lens epithelial cells in different types of age-related cataract.2. The expression level of KCC1 and b FGF had correlation and difference in different types of age-related cataract.3. KCCs and b FGF may take part in the physiologic or pathological metabolic process in the development of age-related cataract. b FGF may affect the expression quantity of KCC1 throgh MAPK/ERK1/2 and PI3K/AKT signaling pathways.
