Objective: To extrapolate the possible mechanism of UTI(Ulinastatin) through analyzing the changes of apoptosis related gene expression profiles of spleen tissue in septic rats, which is under the regulation of UTI preconditioning, based on PCR Assay technology. Methods: Forty-five male Wistar rats were equally divided into sham group, sepsis group and UTI group by means of random number table. In UTI group the rats were treated with intramuscular injection of UTI(100 kUu/kg) an hour before Cecal ligation and puncture(CLP). After surgery,UTI was Administered every 8 hours. In sepsis group and sham group intramuscular balanced solution(5ml/kg) was given. CLP was used to reproduce septic rat model. The sham group was only experienced a simulated operation not CLP. Gene expression profile was studied by using PCR Assay technology to detect the changes in gene expression pattern of rat spleen tissue after CLP. Then using related computer software to screen and analyze the relationship between the Sepsis/UTI group and sham group. At last, analyzing the relation between the Sepsis group and UTI group. Results: During sepsis, abnormal expression of apoptosis-related genes was found, such as in the 96 apoptosis-related genes, there were 8 discrepancy expression between them, including the induction of apoptosis-related genes: 2(Anxa5, Tp63), anti-apoptosis related genes: 3(Cd40lg, Il10, Nol3), Caspase-blockers-associated gene:1(Birc5), cytokine production-associated gene(Lta), prolactin receptor-associated gene 1(Prlr). UTI group, nine appeared differentially experession, including induction of apoptosis-related genes: 2(Bcl2a1, Naip6), anti-apoptosis related genes: 3(Cd40lg, Il10, Naip6), Caspase-blockers-related gene 1(Birc5), cellular electron transport chain-related gene(Cycs), prolactin receptor-related gene(Prlr), housekeeping genes(Ldha). Conclusion: Abnormal expression of apoptosis related genes in the spleen tissue of rats with sepsis owing to immune suppression can be partly relieved under the UTI preconditioning. To some degree,UTI has a protective effect on spleen at the genetic level.
