Objective:Training marrow mesenchymal stem cells of rabbit and poly(hydroxybutytate-co-hyd-roxyoctanoate) osteochondral scaffolds coating with different concentration of gelatin solution together, observe adhesion and proliferation of cell on scaffolds.Methods:Use of solvent casting- particulate leaching method, manufacture poly(hydroxybutyrate-co-hydroxyoctanoate) osteochondral scaffolds with three-layers structure, layer of bone/bone and cartilage interface layer/layer of cartilage,then coating with 3 kinds of different concentration of gelatin solution on the bone layer of the pure material scaffolds. Inject the third generation marrow mesenchymal cells of rabbit on these scaffolds respectively, MTT method is used to detect the proliferation of cells on the scaffolds, then observe cell growth on the scaffolds with electron microscope 10 days later.Results:The integration of bionic scaffolds which manufactured with new materials have three-layers structure, each layer connect compact, uniform pore, space network distributed mutual transportation;Scaffolds of the experimental group were coating with 3kinds of different concentration of gelatin solution, had a higher rate about adhesion and hydrophilic than control group, and significantly increased with the increase of concentration; The cell proliferation activity of the experimental group is higher than that of the control group, concentration of 6% is higher than the other two experimental groupsin the 2th to 6th days, but less than concentration of 4% in the 8th day.Conclusions:Poly(hydroxybutyrate-co-hydroxyoctanoate) osteochondral scaffolds coating with gelatin solution, can increase the adhesion rate of marrow mesenchymal stem cell, and concentration of 4% has a better effect.
