| Articular cartilage plays a very important role in living organisms for bearing load, sliding, shock absorption and other functions, meanwhile the lack of blood vessels and lymphocytes, it leads to a very small self-healing capabilities after injury. Despite many years of clinical researches, the traditional treatment can not achieve a satisfactory results. Cutting operation and debridement just make a secondary damage, at this time new cartilage is prone to degradation, allograft cartilage transplantation’s application is limited for its immune response, autologous cartilage transplantation can not repair larger damage cause their limited sources of material. Finally osteochondral tissue engineering created a new way for repairing the articular osteochondral.Nowadays, the study on joint tissue engineering focuses on stem cell, shelf production, and cell growth factor, especially the shelf production. The previous study on joint tissue engineering mainly investigates the production of cartilage, which ignores the cartilage bone layer. However, cartilage bone layer is important, since it supply nutrition for cartilage. This paper examines the production of cartilage shelf with PHBHOX, the extraction of neonatal rabbit bone marrow stem cell, and the composite culture of shelf and cells.With solvent casting/particulate leaching legal, the shelf could be made. Under the help of chloroform solvent, bone layer-intermediate layer-layer of cartilage would be conected by PHBHOX through pore of these components. The pore apertures are280μm,200μm,≤60μm, respectively, and the corresponding porosity are75%,85%,60%. The Mechanical strength are4.23Mpa,2.75Mpa,6.30Mpa, which reach the requirements of the organisms on the three components of the tissue engineering.In this paper, the seed cells are obtained by bone marrow stem cells (BMSCs) of rabbit marrow, which is purified by adherent method. After72hours of culture, adherence rate reaches60%. Six days later, cells cover the bottom and performance the most popular growth state, which replies the stability of cells and owns high biological activity.After preparation of the stent and seed cells, with a composit culture of them, adhesion rate will be76%24hours later. The cell morphology performs well, which demonstrates the good stent biocompatibility of integrated shelf.In the experiment, BMSCs of rabbit has been obtained and cultured with integrated shelf. Proprietary material of PHBHOX has been adopted to make the integrated shelf of bone layer-intermediate layer-layer of cartilage, with the objective of articular bone and cartilage damage repair. Hydroxyapatite is incoperated into the bone, which supplies the growth nutrition of bone cells and guarantees high adhesion rate. The investigation in this paper provide theorem and experiment basis for further study on joint damage repair with integarated shelf. |
