Objective:The rat model of non-alcoholic fatty liver disease(NAFLD) is bulid to observe the changing of adiponectin(ADPN) and chemokine-like receptor 1(CMKLR1), and to investigate the effect and mechanism of action of glycine on adiponectin and CMKLR1 with NAFLD which can provide new ideas to pathogenesis of clinical research NAFLD and work of diagnosis and treatment.Methods:After the rats were fed adaptively for 1 week, 36 male SD rats whose age we re in 10---12 were randomly divided into three groups(12 rats each group): 1. Contro l group: rats were fed with normal food. 2. High-fat-fed group: 84%common feed,14%lard, 2%Cholesterol feeding. 3. Glycine group: 79%common food, 14%lard, 2%Cholesterol feeding, 5%glycine feeding. These rats were killed at the end of 8 wee ks and 12 weeks respectively. Measuring weight and liver of rats to calculate liver index; adopting the abdominal aorta collected blood detect tumor necrosis factor-α(T NF-α), Alanine aminotransferase(ALT), Aspartate transaminase(AST), Triglyceride(TG)and adiponectin concentration. Liver were collected for HE staining and histologica l change of liver were observed. Taking the ways of Immunohistochemistry and RTPCR to test the expression of adiponectin and CMKLR1 in protein and gene levels in hepatic.Results:1. The weight and liver index in high-fat-fed group were significantly higher than in control and glycine group(P<0.05); Groups of rats weight and liver index increased with the extension of raising time; 2. The level of ALT, AST, TG and T NF-α in high-fat-fed group significantly increased compared with these in control gr oup and glycine group at the same period, glycine group is lower than high-fat-fed group(P<0.05)during the same period; 3. 8,12 weeks rats serum adiponectin in h igh-fat-fed group is lower than these in control group(P<0.05)the same period,glycine group is higher than high-fat-fed group(P<0.05); 4. The histologic change s in the liver: During each period, hepatic lobular structures is clear in control gr oup. Hepatic sinusoid and hepatic stripe centered around the central vein radiated li ne up to the surrounding. Fat droplets and inflammatory cells are not seen in this group. Sizes of spherical lipid drops appears in the cytoplasm, with the large one c an be full of the whole cell and shove the nucleus to oneside, inflammatory cells i nfiltration is visible in part of these cells. Lesion in glycine group is relieved than it in high-fat-fed group at the same time; 5.Hepatic immunohistochemical detection results: Adiponectin levels in glycine group is higher than that in high-fat-fed gro up, CMKLR1 in glycine group is lower than the high-fat-fed group’s; 6. RT-PCR r esults: Gene expression of hepatic adiponectin in glycine group is higher than it in high-fat-fed group(P<0.05), gene expression of CMKLR1 in glycine group is lo wer than high-fatfat-fed group(P<0.05); 7. Linear correlation analysis results: seru m TNF-α positively correlated correlated with hepatic CMKLR1 m RNA expression(r=0.441,P=0.007); serum adiponectin m RNA negatively correlated with hepatic C MKLR1 m RNA expression(r=-0.527,P=0.001); hepatic adiponectin m RNA negative ly correlated with hepatic CMKLR1 m RNA expression(r=-0.52,P=0.001).Conclusions:1. CMKLR1 can increase the degree of inflammation of NAFLD, may play it’s role through inhibiting the expression of adiponectin. 2. Glycine may act it’s liver protection role by downregulate expression of CMKLR1 and upregulate the express ion of adiponectin.
