| Objective:Type 1 diabetic rats model was constructed, the different course of diabetes rat vascular pathological changes, and serum adiponectin, aortic transforming growth factor beta 1, â…£ type collagen protein expression changes, so as to improve the understanding of the pathogenesis of type 1 diabetes vascular, for type 1 diabetes to provide new train of thought and basis of diagnosis and treatment of large blood vessels.Methods:Selected 24, 8 weeks of male SD rats given high-dose chain urea with intraperitoneal injection of cephalosporins in type 1 diabetes model was established, and a normal control group 16.After the success of the building, randomly selected from diabetes group of 20,high-fat feeding;Normal control group 16, ordinary feed;In 4 weeks, 8 weekend between groups randomly selected from 8 only to be put to death.All rats to be put to death before weighing, adiponectin blood test, blood sugar, low density lipoprotein cholesterol,specimens, return the thoracic aorta do HE staining, using immunohistochemical method and RT-PCR,Western blot method to determine the expression of TGF- beta 1, â…£- C.Results:1.General situation: diabetes group rats compared with the corresponding control group hair dry, dark color, depressed, sluggish, the activity decreases, and "a little" sanduo symptoms.Compared with normal control group, diabetes group significantly increased blood sugar, low density lipoprotein cholesterol, weight, adiponectin reduced significantly,the difference was statistically significant(P < 0.01);Compared with diabetes group 4weeks, 8 weeks group of low density lipoprotein cholesterol, diabetes adiponectin is reduced, the difference was statistically significant(P < 0.05).2.HE staining: normal control group in the arterial wall lining, membrane, the outer membrane of three layers structure integrity, newspeak assumes the circular, the wall is smooth.Membrane smooth muscle cells in endometrium flat, size, shape is normal,neatly;Outer membrane normal form.Diabetes group at 4 weeks can be seen in the arterial wall lining, membranes, the outer membrane boundary is not clear, mild disorder,membrane integrity, endothelial cell parts fall off;Membrane smooth muscle cells in size,shape change, cell number increase, disordered arrangement.Diabetes group at 8 weeks the further development of the pathological changes, can be seen in the film structure is loose,interstitial collagen fiber hyperplasia, also have a small amount of foam cell aggregation,lipid deposition, foam cell size can be seen in the cytoplasm of cavitation.3.Immunohistochemical method and RT-PCR,Western blot method are: diabetes group compared with the corresponding control group, the TGF- beta 1, â…£- C levels;Compared with diabetes 4 weeks group, diabetes group 8 weeks TGF- beta 1, â…£-C levels, the difference was statistically significant(P < 0.05).4.Correlation analysis: adiponectin and transforming growth factor beta 1, â…£ collagen are negatively correlated, and transforming growth factor beta 1 and â…£ type collagen were positively correlated, the difference was statistically significant(P < 0.05).Conclusions:1.Adiponectin levels in rats with type 1 diabetes group decreased, with the extension of the course to reduce more significantly, can delay the development of the diabetes vascular lesions.2.Type 1 diabetes group rats transforming growth factor beta 1, â…£ collagen protein expression increased, more apparent with the progression, consistent with the degree of diabetes vascular lesions, is one of the risk factors for diabetes vascular lesions.3.Adiponectin and transforming growth factor beta 1, â…£ collagen were negatively correlated, transforming growth factor beta 1 and â…£ type collagen were positively correlated. |
