Effect Of Deferiprone And Clioquinol On Brain Damage Following Cerebral Hemorrhage In Experimental Rats

Objective:To observe the effect of deferiprone and clioquinol on Hepcidin、Hephaestin、iron contents、reactive oxygen species and functional outcome after intracerebral hemorrhage in rats.Methods:96 male wistar rats were randomly divided into four groups(24 per group), sham operation group 、 intracerebral hemorrhage group 、 deferiprone treatment group and clioquinol treatment group(n=6). Separately observed at neurobehavioral deficit scores;total iron contents by flame atomic absorption spectrometry; reactive oxygen species contents with chemiluminescence method; expression of Hepcidin and Hephaestin by immunohistochemical method at 1d, 3d, 7d and 14 d after operation. All data were analyzed by SPSS13.0.Results:Contrast to sham, iron contents were increased in intracerebral hemorrhage,deferiprone and clioquinol group at each time point. It reached a plateau at day 7 in intracerebral hemorrhage; Contrast to the intracerebral hemorrhage group, iron concents were decreased at 3d, 7d,14 d in deferiprone and clioquinol group. Contrast to sham, reative oxygen species were increased in intracerebral hemorrhage、deferiprone and clioquinol group at each time point. It reached a plateau at day 7 in ICH. There were no significancebetween intracerebral hemorrhage and deferiprone group. Contrast to intracerebral hemorrhage, reative oxygen species were decreased at each time point in clioquinol.Contrast to sham, the expression of Hephaestin were decreased at 1d、3d、7d、14d in intracerebral hemorrhage 、 deferiprone and clioquinol. Different from Hephaestin, the expression of Hepcidin were increased in intracerebral hemorrhage 、 deferiprone and clioquinol at each time point. Contrast to intracerebral hemorrhage, the expression of Hepcidin was dicreased in 3d,and there was no difference between intracerebral hemorrhage and deferiprone in the expression of Hephaestin.Contrast to intracerebral hemorrhage, there were an decrease of Hepcidin at each time point but increase of Hephaestin in clioquinol at every time point. Contrast to sham, the neurological function scores were increased signicantly in intracerebral hemorrhage、deferiprone and clioquinol group at 1d,3d,7d,14 d. It reached a plateau at day 3 in intracerebral hemorrhage 、deferiprone and clioquinol. There was no difference between intracerebral hemorrhage and deferiprone group at each time point. Contrast to intracerebral hemorrhage, there was decrease of neurological function score at 7d、14d in clioquinol.Conclusion:There were an increase of the production of iron contents、reactive oxygen species and the expression of Hepcidin but decrease of Hephaestin after rat intracerebral hemorrhage. deferiprone have no effect on reactive oxygen species and the expression of Hepcidin and Hephaestin and fail to improve the outcome in intracerebral hemorrhage,but can reduce iron overload after intracerebral hemorrhage. clioquinol can decrease iron contents 、 reactive oxygen species and the expression of Hepcidin and increase Hephaestin,then improve the outcome in intracerebral hemorrhage.
