Objective Detecting the expression of Let-7b and proteins translated by its target gene in tissue of thyroid papillary carcinoma(PTC) with/without Hashimoto’s disease(HT) and nodular goiter to shed light on the investigation of Let-7b’s function in the occurrence of thyroid papillary carcinoma with Hashimoto’s disease.Method Extracting micro RNAs from fresh frozen tissue of enrolled patients.Using real-time PCR to detect the expression level of Let-7b in the tissues of PTC patients with/without HT and nodular goiter. Western blot was applied to detect the expression of CCR7 and CCL7 in those patients tissues and Immunohistochemical staining was used to further confirm the result with formalin fixed paraffin embedded tissue. Collecting the clinical data of those patients whose tissue was enrolled for those detection. Comparing the expression of Let-7b, CCR7, CCL7, PTAFR in tissues of three different disease.Investigate the correlation between the expression of Let-7b and CCR7, CCL7, PTAFR and patients clinical characteristics.Result ①The expression of Let-7b was much higher in PTC patients with/without HT than patients with nodular goiter. After calculation, the result wasΔΔCt=ΔCt PTC-ΔCtnodular goiter= 1.25,2-ΔΔCt =0.61,ΔΔCt=ΔCt PTC with HT-ΔCtnodular goiter =2.02,2-ΔΔCt =0.38, which indicates that the expression level of Let-7b in PTC patients with/without HT was 0.61 and 0.38 times less than that in nodular goiter patients respectively. The expression level of Let-7b in PTC with HT patients was higher than that in PTC patients without HT, After calculation, the result was ΔΔCt=ΔCt PTC with HT-ΔCt PTC=0.77, 2-ΔΔCt =0.62, which means that the expression level of Let-7b in PTC with HT patients was 0.62 times less than that in PTC patients without HT, however this difference was not statistically significant. ②CCR7 was mainly located in cytoplasm and cytolemma. The expression of it in PTC with HT patients was the highest, following that in PTC patients. The CCR7 expression in nodular goiter was the lowest. The expression of CCR7 was related to the serum level of FT3. ③ CCL7 was mainly located in cytoplasm and cytolemma, too. The expression of it in PTC with HT patients was the highest followed by that in PTC patients. The CCL7 expression intissues from those two groups was much higher than that from nodular goiter. It’s expression was related to the rate of lymph node metastasis(r=0.573,P<0.01)。④PTAFR was mainly located in cytoplasm and cytolemma, too. The expression of it in PTC patients was slightly over that in. PTC with HT patients. Both the expression of Let-7b in those two tissues were more than that in HT patients. It’s expression in nodular goiter was the lowest.Conclusion Low expression of Let-7b plays a role in the development of PTC in HT patients, which might be achieved through the regulation of the transcription levels of CCR7、CCL7 and PTAFR.
