OBJECTIVE To study the cytotoxicity of Graphene-polyethylene glycol-doxorubicin(GO-PEG-DOX) using lymphoma cells model which will provide a basis for the clinical application of nanodrug instruction purpose of clinical medication.METHODS First, GO-PEG was successfully prepared by chemical methods, then graphene-PEG loading with DOX was obtained GO-PEG-DOX. The transmission electron microscopy(TEM) was used to characterize the morphology and structure of GO.Functional groups of GO-PEG were analyzed by Fourier transform infrared spectroscopy(FTIR) spectra. UV-Vis was used to characterize of GO-PEG-DOX. Second, lymphoma cells were treated with different concentrations(0-100 μg/m L) of GO-PEG/GO-PEG-DOX at different time points(6, 12, and 24 h). The cytotoxicity of GO-PEG/GO-PEG-DOX on human lymphoma cells was assessed by the CCK-8 assay.The cell apoptosis of GO-PEG/GO-PEG-DOX on lymphoma cells( Raji cells) were assessed by Annexin V-FITC method.RESULTS GO-PEG was successfully prepared by chemical methods。And experient data indicates that GO-PEG has good solubility in water, 0.9% Nacl, PBS and RPMI-1640 medium. The results indicate that the cytotoxity of GO-PEG was time-dependent and does-dependent. The relation of cytotoxicity of GO-PEG on lymphoma cells both concentration of complex and time were positive correlation. The experimental group and the control group compared to the defference was statistically significant(P<0.05). The results showed there was significant defference in cell viability between GO-PEG-DOX group and DOX group(p=0.02), but removing the impact of the cell viability of GO-PEG,there was no significant defference in cell viability between GO-PEG-DOX group and DOX group(P>0.05), indicating that GO-PEG did not decrease DOX cytotoxicity. There Was significant difference between DOX group and control group,and GO-PEG-DOX group and control group(P<0.05). There Was no significant difference between the GO-PEG-DOX group and DOX group(P>0.05).CONCLUSIONS The toxicity of GO-PEG to lymphoma cells was low, and GO-PEG did not decrease DOX cytotoxicity. The results indicated that DOX could induce cellapoptosis, and GO-PEG did not induce apoptosis. The results also indicated that GO-PEG did not change the ability of DOX to induce apoptosis.
