The Apoptosis Influence Of Chitooligosaccharides On AGS Cells Infected By Helicobacter Pylori

Aims We investigate the influences of Chitooligosaccharides(COS) on Helicobacter pylori(HP) growth, and the apoptosis effects of COS on cultured AGS cells and HP infected AGS cells, to discuss the medicinal value of COS as medicinal prodrugs.Methods1. We added COS to the BHI medium on which HP was routinely cultured for 48 Hs,and HP were incubated for another 48 Hs, bacterial colonies were observed under microscope.2. We created HP-AGS cells co-culture model with multiplicity of infection was200.We diluted COS at the proportion of 0.5:100, 1:100, 2:100 and 4:100.Added deferent concentrations of COS to AGS cells and HP infected AGS cells to incubate for 24 Hs in a37℃, 5% CO2 humidified incubator.3. We added Bi Cl3 solution at the concentration of 100mol/ L、200mol/ L,300mol/L and 400mol/ L together with different proportion dilution COS to AGS cells.Cells was incubate for 24 Hs in a 37℃, 5% CO2 humidified incubator.4. The cells apoptosis of AGS cells were analyzed by using Hoechst 33258 stain under fluorescence microscopy.Results1. HP colonies added with COS were apparently decreased compared with the untreated group.2. Hoechst 33258 staining revealed that with the increase of COS concentration,the proportion of highlight staining AGS cells increased, while the normal blue stain cells proportion decreased. In HP infected group, with low concentration of COS, the proportion of highlight staining cells were decreased compared with no COS.3. We found that AGS cells staining was strong stained when added COS together with Bi Cl3.Conclusion1. Chit oligosaccharides may suppress the growth of HP.2. COS induced apoptosis of AGS cells in a dose dependent manner, while Bi3+can significantly enhance its inhibition efficacy.3. COS may weaken the apoptosis of AGS cells induced by HP infected at a low,as which COS protect AGS cells infected by HP at certain concentration.
