This experiment studied Water-soluble polysaccharide in Adenophora wilsonii using Adenophora wilsonii as material, determined and compared the content of polysaccharide in every part of Adenophora wilsonii and the whole plant. In addition, the extraction process of Water-soluble polysaccharide in Adenophora wilsonii was optimized, and the monosaccharide composition of Adenophora wilsonii was determined. Finally, the Water-soluble polysaccharide in Adenophora wilsonii were classified and chemically analized. Meanwhile, their antioxidant activity was also studied. The results were as follows:1. Four extraction methods could be sorted according to rate from high to low as followed: Boiling method>Enzymatic method>Microwave method > Ultrasonic method. It suggest that Boiling method is the most optimal method to extract Water-soluble polysaccharide from Adenophora wilsonii. Additionaly, the determined content of polysaccharide in the root, stem and leaf of Adenophora wilsonii were 3.32%、3.47 ' 3.48% respectively using the extraction methods of Boiling method. No significant difference between them suggest that Water-soluble polysaccharide were evenly distributed in Adenophora wilsonii.2. Single factor and L9(34) orthogonal experiment were cattied out to study the extraction methods for Water-soluble polysaccharide in Adenophora wilsonii using the receiving rate of Water-soluble polysaccharide, and its optimized extraction process was established. The best time for extraction is 4h, the tempreture is 95℃, the times of extraction is three and the ratio of material to liquor is 1:30。 3. After sugar nitrile acetic deriving and GC-MS analyzing, the result can be get that Adenophora wilsonii is composed of five kinds of monosaccharide including rhamnose, arabinose, xylose, mannose and glucose.4. Na Cl solution of different concentrations divided polysaccharide in Adenophora wilsonii into four levels: SWP1,SWP2,SWP3 and SWP4. And the antioxidant activity of different components can be sorted from high to low as followed: SWP2>SWP3 > SWP1>SWP4>SWP Congo red experiments occurred redshift, indicating that the presence of Poly Triple Helix structure Adenophora wilsonii.
