Objective This study through the observation of the main effective components of angelica extract and separation of the extract to be vascular dementia rats nerve the Bcl-2, and the influence of Bax apoptosis cells, from the resistance to apoptosis of cerebral protection mechanism to explore the mechanism of treatment of VCI provides the experimental basis for the clinical treatment of vascular dementia.Method The experiment adopts the bilateral common carotid artery model of vascular dementia ratsrepeatedly clipping method of closed plus unilateral common carotid artery ligation system. 60 male wistar rats were randomly divided into sham-operation group(S), model group(M), nimodipine group(N), Angelica naphtha group(V), Angelica polysaccharides group(P), Nimodipine and Min Angelica sinensis group(X)six groups.The micien group sham operation group,only issolated vascular ligation of artery,no injection of sodium.nitroprusside. The rest of the M, N, V, P, X in each group repeated occlusion of bilateral common carotid artery occlusion of blood flow with unilateral carotid artery ligation and injection of sodium nitroprusside, preparation of chronic cerebral hypoperfusion in rats with vascular cognitive impairment model. S, M group were givensaline, N group was given nimodipine gavage, V group was given the volatile oil by gavage,group P treated with Angelica Polysaccharide by gavage, X group was given nimodipine and Angelica sinensis gavage,Once per day, continuous lavage treatment of 4weeks.Behavioral experiments(water maze method)carried out after 4 weeks. After the end of the experiment behavior,the rats were decapitated, specimens of brain tissue, the choice of the hippocampus, specimens were fixed and sliced, for HE staining, the expression of Bcl-2 was detected by immunohistochemistry and Bax, a high resolution color pathological image analysis system,statistical data. With angelica root essential oil, angelica polysaccharide group and nim horizontal comparison, observation he angelica to be VCI group rats the Bcl-2 and Bax expression effect.Results 1. through repeated bilateral common carotid artery ligation of VCImodel is successful, the incubation period model group rats were significantly longer than control group.2. the Morris water maze experiment shows: the intervention group have im proved the learning and memory ability, Angelica sinensis + nimodipine group me mory is better than nimodipine group, angelica polysaccharide group and Angelic a volatile oil group.(P<0.01), the difference was statistically significant;3. immunohistochemical results show: the results of immunohistochemistry showed that Angelica sinensis + nimodipine group reduce cell apoptosis in rat VCI trend, protect nerve cells.4.nimodipine group, angelica polysaccharide group, angelica volatile oil group, and Angelica sinensis and nimodipine group were able to promote the expression of Bcl-2 protein in rat model of VCI(P<0.05), and Angelica sinensis + nimodipine group has the best effect(P<0.01), the difference was statistically significant.5.nimodipine group, angelica polysaccharide group, the expression of Angelica volatile oil group, and Angelica sinensis + nimodipine group were able to inhibit Bax protein in rat model of VCI(P<0.05), and Angelica sinensis + nimodipine group decreased significantly(P<0.01), there are significant statistical difference.Conclusion Angelica sinensis can reduce the expression of vascular cognitive impairment model of apoptosis in brain tissue of rat Bax gene, increased expression of Bcl-2 gene with neuronprotective effect, thereby inhibiting the apoptosis of nerve cells, accelerate the recovery of nerve function, together play a role in brain protection.
