Study On The Mechanism Of Regulating The Expression Of Apoptosis Related Proteins And Improving The Function Of Cognitive Dysfunction By Huan Chi Capsule

Purpose: In this study,VCI model rats were made by bilateral common carotid artery ligation(2VO).The behaviors of the apoptosis-related proteins in hippocampus with VCI model rats were observed and the changes of the index of the enzyme.Using traditional Chinese medicine theory to guide and explore the treatment of VCI disease ideas and the basis of Huanchi capsule.Method: SD rats were modeled by 2VO method and randomly divided into 6 groups(n = 11).They are the model group,the sham operation group and the positive control group.The positive control group were divided into three groups: high,middle and low.Behavioral testing was performed by electrical stimulation(jumping method).Biochemical markers were detected by enzyme-linked immunoassay(ELISA).Result: The results of Behavior showed that comparing with the model control group,the disease level of the rats was significantly lower than that of the model control group(P <0.05 or P <0.01).Compared with the model control group,the results of ELISA showed that the HUANCHI capsule had a certain inhibitory effect on the expression of A?1-40 protein,P53 protein and Bax protein in the hippocampus of model rats(P <0.05 or P <0.01),and the activity of Caspase-3 was decreased(P <0.05 or P <0.01)and the expression of Bcl-2 protein was increased(P <0.05 or P <0.01).Compared with the model group,HUANCHI capsule may reduce the Bax protein and Bcl-2 protein ratio.Conclusion: The results of behavioral experiments show that the Huanchi capsule can significantly improve the learning and memory ability of the model rats,prolong the incubation period of the model animals for the first time jumping off the platform and reduce the number of errors.The results of ELISA show that the HUANCHI capsule could inhibit the expression of protein or the activity of the protein in the hippocampus of the model rats which could increase the expression of the apoptotic protein.As well as to increase the expression of antagonistic cell apoptosis protein.It is suggested that HUANCHI capsule could improve VCI symptoms may be related to its involvement in the process of apoptosis.
