Objective: To investigate the effect and mechanism of raw atractylodes on preventing and treating gastrointestinal dysfunction induced by the acute mountain invironment.Method: 70 Wistar rats were randomized into 7 groups, control group(lanzhou altitude,1430 m, group A), high altitude model group( 3500 m, group B1; 5000 m, group B2), maleic acid trimebutine group(3500 m, group C1; 5000 m, group C2), and raw atractylodes group(3500 m, group D1, 5000 m, group D2), 10 rats in each group. Except the rats of group A, the remaining rats were fed 3 days at their own altitude in the low pressure oxygen tank, in which the rats of group A and group B were given saline lavage, and the rats of group C were given maleic acid trimebutine lavage, while the rats of group D were given raw atractylodes lavage.After 3 days, all rats were fed 2 m L ink lavage and sacrificed after 30 min, after that to measure small intestine propulsion rate of ink. The gastrointestinal mucosa pathological damages were observed by HE staining. The m RNA’s expression of proto-oncogene C-kit was measured by q PCR. The expression of serotonin 4 receptor(5-HT4) and dopamine D2receptor(D2DR) in Interstitial cells of Cajal(ICC) of gastric antrum tissue and small intestine tissue were detected by immunohistochemical method.Results:1. The small intestine propulsion rate: Compared with group A, group B2 was significantly decreased(P<0.05), and group B1 was significantly increased(P<0.05);Compared with group B1, group C1 had no significant difference(P>0.05), and group D1 was significantly increased(P<0.05); Compared with group B2, group C2 and D2 were significantly decreased(P<0.05); Compared with group A, there were some differences in group C1(P<0.05), and there were no obvious differences among group C2, D1, and group D2(P>0.05).2. Gastrointestinal tissue pathological damage: The gastric mucosa tissues were normal in group A. The pathological damages in gastrointestinal mucosa tissues were moderate in group B1. The pathological damages in gastrointestinal mucosa tissues were serious in group B2.There were significant differences among the three groups(P<0.05); Compared with group B1,the pathological damages in gastrointestinal mucosa of group C1 and D1 were significantly decreased(P<0.05), while there was no obvious difference between group D1 and group A(P>0.05); And group B2 and C2 had no significant difference(P>0.05). The pathological damages in gastric mucosa were significantly decreased in group D2(P<0.05), but still had difference from group A(P<0.05).3. The m RNA’s expression of proto-oncogene C-kit: compared with group A, group B1 was obviously decreased(P<0.05), and group B2 was obviously increased(P<0.05);Compared with group B1, group C1 and D1 were obviously increased(P<0.05), Compared with group B2, group C2 and D2 were significantly decreased(P<0.05); Compared with group A, there were differences in group C1(P<0.05), and group C1, D1, and group D2 had no obvious difference(P>0.05).4. The positive expression of ICC, 5-HT4, D2DR: Compared with group A, the positive expression of ICC, 5-HT4 in group B1 was significantly decreased(P<0.05), and the positive expression of D2 DR was significantly increased(P<0.05); the positive expression of ICC,5-HT4 in group B2 was obviously increased(P<0.05), the positive expression of D2 DR was significantly decreased(P<0.05). Compared with group B1, the positive expression of ICC,5-HT4 and D2 DR in group C1 had no obvious difference(P>0.05), while the positive expression of ICC, 5-HT4 in group D1 was obviously increased(P<0.05), and the positive expression of D2 DR was significantly decreased(P<0.05). Compared with group B2, the positive expression of ICC, 5-HT4 in group C2 and D2 was significantly reduced(P<0.05), the positive expression of D2 DR was significantly increased(P<0.05); Compared with group A,the positive expression of ICC, 5-HT4, and D2 DR in group C1 had difference(P<0.05), while group C2, D1, and D2 had no obvious difference(P>0.05).Conclution:1. The acute mountain environment leads to gastrointestinal dysfunction, which is closely related to altitude,and the mechanism is related to the change of ICC、C-kit、5-HT4、D2DR in the gastrointestinal tissues.2. The acute mountain environment leads to the pathological damage of gastrointestinal tissues. The higher the altitude, the damage of tissue is heavier.3. The raw atractylodes plays a role in protecting the gastrointestinal mucosa andpreventing and treating gastrointestinal dysfunction induced by the acute mountain invironment through regulating the expression of ICC、C-kit、5-HT4、D2DR.
