Objective: The gastrointestinal nerve growth factor(NGF),stem cell factor(SCF)and estrogen ? receptor(ER?)of low pressure hypoxia Wistar rat model were treated with Atractylodes macrocephala.To observe the effect of hypobaric hypoxia on the expression of NGF,SCF and ER? in gastrointestinal tract of rats,and to study the effect of Atractylodes macrocephala Koidz on the expression of NGF,SCF and ER?in gastrointestinal tract of hypoxic rats.To explore the correlation between SCF,ER?and C-kit and verify the effectiveness of Atractylodes macrocephala on gastrointestinal motility dysfunction induced by hypobaric hypoxia.Method: A total of 70 SPF Wistar rats were randomly divided into 7 groups:blank control group(Lanzhou altitude,1520 m,group A),low oxygen chamber model group(altitude 3500 m,group B1),low oxygen chamber model group(altitude 5000 m,group B2),low oxygen chamber raw white group intervention group(altitude 3500 m,group C1),low-pressure oxygen chamber Health and Atractylodes intervention group(altitude 5000 m,group C2),hypoxia chamber positive control group(altitude 3500 m,group D1),low-pressure oxygen chamber positive control group(altitude 5000 m,group D2),each group of 10 rats.Group A rats were placed in animal breeding room,group B1,C1,D1 placed in the simulated low pressure hypoxia 3500 m altitude oxygen chamber,group B2,C2,D2 placed in the simulated low pressure hypoxia 5000 m altitude oxygen chamber,feeding time is 3d(72h),group A and group B1 and group B2 were given normal saline for gavage,group C1 and C2 were given algae solution for gavage,group D1 and D2 were given tricameline maleate solution for gavage.The expression of SCF,ER? and C-kit were detected by western blot in immunohistochemistry,the expression of NGF was detected by immunohistochemistry.Results:1.NGF positive area expression: compared with group A,the positive area of group B1 was decreased(P<0.05),and the positive area of group B2 was increased(P<0.05),compared with group B2,the positive area of C2 group and D2 group wasdown-regulated(P<0.05),compared with group D1,the positive area of group C1 was increased(P<0.05),compared with D2 group,there was no significant difference in the positive area of C2 group(P>0.05).2.SCF protein expression: compared with group A,the expression level of group B1 was decreased(P<0.05),and the protein expression level of group B2 was increased(P<0.05),group D1 and group D2 showed a decreasing trend(P<0.05),there was no significant difference between the C1 group and the C2 group(P>0.05),compared with B1 group,the expression level of group C1 and group D1 were up-regulated(P<0.05),compared with B2 group,group C2 and group D2 protein expression were down-regulated(P<0.05),compared with group D1,the group of C1 protein expression was increased(P<0.05),compared with group D2,there was no significant difference in the expression level of group C2(P>0.05).3.ER? protein expression: compared with group A,the expression level of group B1 was decreased(P<0.05),and the protein expression level of group B2 was increased(P<0.05),group D1 and group D2 showed a decreasing trend(P<0.05),there was no significant difference in the expression of group C1 and C2(P>0.05),compared with B1 group,the expression level of group C1 and group D1 were up-regulated(P<0.05),compared with B2 group,group C2 and group D2 protein expression were down(P<0.05),compared with group D1,the expression level of group C1 was increased(P<0.05),compared with group D2,there was no significant difference in the expression level of group C2(P>0.05).4.There was a positive correlation between SCF and C-kit protein expression(r = 0.950,P<0.05),a positive correlation between ER? and C-kit protein expression(r = 0.786,P<0.05).Conclution:1.Hypobaric hypoxia may lead to gastrointestinal motor dysfunction,also lead to abnormal expression of NGF,SCF and ER?.2.SCF and ER? regulating gastrointestinal motor function via changing the expression of C-kit and affecting the ICC growth,development and pacing function.3.Raw atractylodes in the low-pressure hypoxia-induced gastrointestinal motordysfunction to intervened by regulating expression of NGF,SCF and ER?,its effect is better than tramadine maleate capsule.
