Objective:In order to provide clinical guiding significance for XELOX neoadjuvant chemotherapy individually in the future,we investigate the correlation between expression levels of ERCC1/TS/TP m RNA and the efficacy of XELOX neoadjuvant chemotherapy for patients with colorectal cancer. Methods:79 fresh fibercoloscope biopsy tissues were obtained from the patients firstly diagnosed with colorectal cancer before chemotherapy and out of these patients,48 cases met inclusion criteria.Only 42 patients received neoadjuvant chemotherapy of three cycle XELOX regime(oxaliplatin/capecitabine) completly before surgery. After chemotherapy,all of the 42 patients had an suegery and we collected these operation specimens.The expression levels of ERCC1/TS/TP m RNA of each biopsy tissue were detected by q RT-PCR. The chemotherapeutic response was evaluated with and Dworak’ TRG Criteria(pathological criteria). The correlation between the expression levels of ERCC1 m RNA,TS m RNA and TP m RNA and the chemotherapeutic efficacy of XELOX regime were analyzed. Results:1.The response rate of neoadjuvant preoperative chemotherapy in this cohort study was 52.38 percent based on Dworak’ TRG Criteria.2.Based on Dworak’ TRG Criteria,the efficacy rate of patients with low expression levels of ERCC1 or TS m RNA were significantly higher than that of patients with high expression levels(P<0.05),the efficacy rate of patients with low expression levels of TP m RNA were significantly lower than that of patients with high expression levels(P<0.05).3.The pathological remission rate of two or three out of ERCC1,TS,TPm RNA was significantly higer than zero or one gene m RNA4. The and pathological remission rate of poorly differentiated patients was lower than that of patients with moderately and well differentiated patients significantly. The difference was statistically significant.(P<0.05). The significantce between effect of chemotherapy and gender, age, tumor gross type, ECOG score of patients were not significant(P>0.05). Conclusion:The expression of ERCC1/TS/TP m RNA can predict efficacy and response in colorectal cancer patients treated with XELOX regime.Combined detection the expression of ERCC1,TS and TP m RNA will provide theoretical basis of individual treatment for patients with colorectal cancer.
