Objective To analyze the profile of hepatitis B virus(HBV) resistance mutations against nucleos(t)ide analogues(NAs) treatment in patients with chronic hepatitis B(CHB) or with HBV-liver cirrhosis for more coprehensive information to assess disease progress and to implement further antiviral treatment step. Methods A total of 147 patients with chronic hepatitis B or HBV-liver cirrhosis after virological breakthrough aganist antiviral nucleos(t)ide treatment from the afflliated hospital of Ningxia Medical University from Janury 2012 to December 2012 were selected. We used genes direct sequencing method to aalyse teir PCR products of serum HBV P gene and the resistant sites and the relevent medical history was reviewed. Results We found the mutation rate was 53.06%,and there were main forms like A181T/V、M204V/I、N236T. The rest were diverse and most of all were united mutant forms. The use of the drug and its generated resistance locies were basically identical. The main mutant sites were A181T/V. LAM-resistant in 28 cases,ADV-resistant in 50 cases,ETV-resistant in 0 cases. And the rest dispersed. Primary drug resistance were drug stopping. Conclusion It plays a very important role in rational drug use and adjusting antiviral treatment that the study of HBV P area mutations of antiviral of chronic hepatitis B and liver cirrhosis patients after virological breakthough.Adherence is an important cause induceing NAs resistance in CHB patients. The ciinicians should attention to the patients with NAs teratment for reducing the risk of drug resistance.
