Objective: It will detect the expression of serum and glucocorticoid-regulated kinase 1(SGK1) in precancerous lesion and esophageal cancer tissue by using immunohistochemical, Western blotting and real-time quantitative RT-PCR, to explore the role and significance of SGK1 protein in the development of esophageal cancer, and provide evidence for the early diagnosis of esophageal cancer, looking for possible interventions and treatment method.Methods: Collected the 60 pathological diagnosis cases with esophageal cancer resection from January 2011 to October 2012 at the First Affiliated Hospital of Henan University of Science and Technology, and took the cancer tissues and the corresponding adjacent tissues. Respectively, using immunohistochemistry, real-time quantitative RT-PCR and Western blotting were performed to detect three methods SGK1 protein in esophageal carcinoma, adjacent tissues and normal esophageal mucosa situation. Applied SPASS17.0 statistical software to analyze the relation between the expression of SGK1 and esophageal carcinoma patients with specific clinical pathological types and tissue samples, and the survival of patients among different SGK1 expression were analyzed.Results:1. Immunohistochemistry results showed that the positive rate of SGK1 protein in esophageal carcinoma tissues and the corresponding adjacent tissues and normal esophageal mucosa tissues were 65%, 36.7%. The expression of SGK1 in esophageal carcinoma and were increased compared with in the corresponding adjacent tissues, there are statistical differences(P < 0.05). It was analyzed that SGK1 positive expression in tumor differentiation, depth of invasion, lymph node metastasis and clinical stage had significant difference(P < 0.05, P < 0.05, P < 0.05, P < 0.05).2. Western blotting results showed that the SGK1 protein has significantly higher expression in esophageal carcinoma tissues than in the corresponding adjacent tissues, and there are significant differences(P <0.05).3. RT-PCR results showed that the expression of esophageal carcinoma tissues was obviously higher than the corresponding adjacent tissues in esophageal cancer specimens, and had significant difference(P < 0.05).4. Correlativity of the results of Western blotting and RT-PCR were analyzed, the results revealed a positive correlation, and the significant correlation.It showed that the expression of SGK1 protein and mRNA was consistent.Conclusion:1. The expression of SGK1 was closely related with the esophageal carcinoma invasion depth, differentiation degree, lymph node metastasis and clinical stage. It suggested that SGK1 was related with the malignant phenotype and invasive ability of esophageal carcinoma.2. There was significant difference of SGK1 expression in esophageal cancer tissue, corresponding adjacent tissues. Conclusion One indicated that the SGK1 may related to the occurrence and progress of esophageal carcinoma.
