Objective: Analysis the differences of the serum biochemical changes between the healthy control group and the patients with benign thyroid tumor 2 months and 4 months postoperatively, to demonstrate if different surgical method with taking levothyroxine tablets could cause different effects for thyroid benign tumor patients in metabolomics levels.Methods: 1. According to the exclusion criteria,12 cases of the patients after total thyroidectomy(T0 group), 20 cases of the ones after hemi thyroidectomy(H0 group) were collected from the Union Hospital affiliated to Fu Jian Medical University in November 2013 to October 2014. Then collect their serum specimen 2 months and 4 months postoperatively. At the same period, collecting the serum specimen of 20 cases volunteers from medical examination center(C group).All the serum specimens were stored in eighty degree below zero. 2. Analysis the serum specimen of above sixty patients with Switzerland Bruker 500 Hz NMR. 3. To phase and baseline correction use Mest Re Nova(7.0) software,to integral between 0.0-0.5 ppm after calibration of the spectra,the width is 0.002 ppm. 4. Analysis the differences about endogenous metabolites between the treatment group and control group at each time point by PCA and PLS-DA with SIMCA-P+.Results: 1. The data between T group and C group has good clustering in PCA., PLS-DA discrimination model is established with good reliability and predictability(T1 Vs C R2X=0.871 Q2X=0.830; T2 Vs C R2X=0.879 Q2X=0.838). Significant metabolites changes is judged by OPLS-DA analysis and Pearson’s product-moment correlation coefficient. Compared with the healthy control group, the content of metabolites in theserum of total thyroidectomy group are reduced,including leucine, isoleucine, valine, arginine, NAG, glutamate, pyruvate, glutamine,creatine,choline,PC,GPC, lactate, histidine, R-CH3, while the content of Glucose are increased. 2.The data between H group and C group has good clustering in PCA., PLS-DA discrimination model is established with good reliability and predictability(H1 Vs C R2X=0.889 Q2X=0.864; H2 Vs C R2X=0.860 Q2X=0.820). Significant metabolites changes is judged by OPLS-DA analysis and Pearson’s product-moment correlation coefficient. Compared with the healthy control group, the content of metabolites in the serum of total thyroidectomy group are reduced,including leucine, isoleucine, valine, arginine, NAG, glutamate, pyruvate, glutamine,creatine,choline,scyllo-inositol, lactate, histidine, R-CH3, while the content of Glucose are increased. 3.The data between T group and H group has poor clustering in PCA.,although the discrimination model has poor Reliability and predictability,we try to find out the significant metabolites changes by OPLS- DA analysis and Pearson’s product-moment correlation coefficient. Compared with H1 group, the content of metabolites in the serum of total thyroidectomy group(T1 group) are reduced,including lactate, phenylalanine, valine, while the content of Glucose are increased. Compared with H2 group, the content of metabolites in the serum of total thyroidectomy group(T1 group) are reduced, including Choline,PC,GPC, R-CH3.And those changes are independent of time.Conclusion: 1. Levothyroxine tablets could not completely replace the function of thyroid in metabolomics levels. 2. There is not enough evidence that different surgical method could cause different effects thyroid benign tumor patients with small sample analysis in a short period.
