Objective To study ubiquitin-specific peptidase 22(ubiquitin specific peptidase 22), which occurs in the kidney tumor tissue about gene and protein expression in tumor tissues and its peri-cancerous normal tissues( kidney tumor ≧distance 2CM),and analyse the expression differences of usp22 between tumor tissues and peri-cancerous normal tissues, combining with clinical and pathological datas. To investigate the possible existence of the causal relationship, then get the correlation significance.Method Analysis were collected from July 2012 to December 2013, the First Affiliated Hospital of Xiamen University,Urology Surgical resection radical nephrectomy specimens of 59 cases of tumor,pathology was reported to confirm malignancy after surgery, lymph node metastasis and pathological data and pathological grading. Corresponding adjacent tissues and 59 cases of various cancer tissues, ≧including cancer adjacent tissues taken from the distal side( 2CM) of cancer tissues, and cancer tissue section was derived cancer tissues, where necrosis did not occur. Including 40 male patients, 19 female patients, around the age between about 30-91 years old, average were 58.2 years. Cancer tissues were obvious the naked eye can identify the cancerous tissues and away from the tumor margin. Patients who undergoing radical nephrectomy prior to surgery were not received any radiotherapy or chemotherapy. The majority of the tissues were clear cell carcinoma. Using Trizol Extraction of the RNA from 20 patients, carcinoma tissues and corresponding beside tissues, then reversed transcription and synthesis of specific primers for real-time quantitative polymerase chain reaction(rt-pcr) to detect different cases of USP22 gene expression.By immunohistochemistry(two-step) method to detect all specimens usp22 protein; extraction of 20 of theirs protein between carcinoma and peri-cancerous normal tissues do testing. Combine with clinical data, the differences were observed between cancer tissues and normal tissues. Correlation analysis and comparative analysis of adjacent tissue and cancer tissue differences in RNA and protein levels of ubiquitin specific peptidase 22.Results In 59 cases of kidney tumor specimens, renal clear cell carcinoma accounted for 51 cases(86.4%), papil ary carcinoma 3 cases(5.1%), chromophobe cell carcinoma 1 case(1.7%), 1 case of spindle cell carcinoma(1.7%), renal epithelioid angiomyolipomas 1 case(1.7%), acidophilic cell adenoma in 2 cases(3.4%). After the examination, in 59 specimens, immunohistochemical results showed that the group of tissues adjacent to carcinoma, the positive expression rate as high as 93.2%, protein mainly located in the cytoplasm.Cancerous tissue protein expression positive rate of 47.5%, and the expression intensity was relatively weaker than tissues adjacent to carcinoma,the difference was statistically significant.The result of immunohistochemistry was the same as Western Blotting and RT-PCR. The expression of protein and m RNA in cancerous tissue was weaker compared to adjacent tissue(≧ 2 cm)。With reportes about USP22, much literatures on human tumor said that USP22 was higher than normal tissues. Our results does not match other experimental results. Our results show that when renal carcinoma(most of them are clear cell carcinoma) occurs, the expression of USP22 has not increased,on the contrary, compared with tissue adjacent to carcinoma(≧ tumor 2 cm) its expression decreased, the results have statistical significance(P < 0.05)Conculsion When renal cancer( mainly clear cell carcinoma) occurs, the cancer tissues expression level was not higher than the adjacent tissues( kidney tumor distance ≧ 2CM).The ubiquitin-specific peptidase 22 as a potential marker of cancer stem cells in many human tumors when its expression increases, may increase the expression of certain genes in tumor tissue, thereby causing malignant transformation of normal cells, but this experiment indicated that during kidney cancer, its expression did not increase, its mechanism is not clear at present,and requires further research is needed to explain its mechanism.
