Objectives:To study the effect of bone marrow mesenchymal stem cells(BMSCs) transplantation treatment combined with erythropoietin(EPO) on the neurological functional recovery in rats with spinal cord injury and explore the possible molecular mechanism.Methods:BMSCs were isolated and cultured. The rat spinal cord injury model was prepared according to the modified Allen method.75male Wistar rats were randomly divided into a sham-operation group, spinal cord injury(SCI) group, EPO treatment group, BMSCs transplantation gourp and the EPO and BMSCs combined group.5000U/kg rhEPO was peritoneal injected at24h before spinal cord injury. BMSCs were transplanted by tail vein after spinal cord injury. Functional outcome measurements were evaluated by the BBB score and the morphological changes were observed by HE staining at different time points. The protein level of brain derived neurotrophic factor (BDNF) was detected by Western-blot at14d.Results:An obvious improvement in locomotion function recovery and the morphological changes was observed in EPO group or BMSCs group than SCI group. The outcome of the combined goup was the best (P<0.05). The Western-Blot results showed that the BDNF expression level of the sham-operation group was the lowest among the five groups, while it was slightly increased in the SCI group. Compared to these two groups, the expression of BDNF obviously increased in EPO group and BMSCs group. It was even higher in the combined group than that in EPO group or BMSCs group.Conclusion:BMSCs transplantation treatment combined with EPO can obviously recover the neurological function in rats with spinal cord injury by up-regulation the expression of BDNF.
