Objective:ADAM is a disintegrin metalloproteinase(a disintegrin and metalloproteinase) family, ADAMs was be found in a lots of plants, animal, biological and even human’s tissues and organs, some studies have demonstrated that human brain, liv-er, lung, heart, ovary, testis, colon, breast and placenta, has ADAMs distribution[1]. It has been more than 40 kinds of ADAMs was be found, such characteristics as the type zinc containing protease general family of transmembrane proteins, the interaction between cell and cell, cell and matrix plays a regulatory importan t role[2]. Research has shown that a disintegrin and metalloproteinase 15(ADAM 15) in tumor metastasis plays an important role [3]. Through the analysis of AD AM 15 in non tumor tissue of breast cancer, breast cancer, metastasis of lymph nodein the expression, discuss the relationship between features and clinical index and pathology in patients with E-cadherin and the invasion and metastasis of bre-ast cancer,biological target for the study of treatment of breast cancer to provide experimental basis for treatment.Methods:(1) cases selection and screening of samples: Paraffin embedded specimens from 60 patients with breast cancer(Through theoperation after resection of pathological diagnosis of breast cancer, and in pati-ents without preoperative chemotherapy and the other for the treatment of breast cancer tissue specimenMetastatic lymph nodes in 23 cases(Breast tissue specimens and lymph-node specimens for the same patients) 30 cases of non tumor tissue of breast(2) Using immunohistochemical technique(SP method) for detection of ADAM l5,E-cadherin in tissues and non tumor breast tumor tissue, pathological diag-nosis is the expression of the metastasis of lymph node in breast cancer. According to the Fromowitz score standard statistical data.(3) Application of SPSS l7.0 software and statistical analysis of the experimental data was, according to the analysis of different types of indexes was used the 2 test or Fisher exact test, rank transformation of non parameter test and Spearman correlation analysis; test standards to P<0.05 for statistical significance.Result:In breast non tumor tissue,ADAM 15 has Low expression. In breast cancer tissues and metastatic lymph nodes ADAM l5 has high expression. Its expression had no correlation with patient age, menstruation, the size of the tumor can cause the change of ADAM l5 expression, the expression degree and clinical ADAM l5 staging, tissue malignancy grading is proportional to the. Expression of E-cadhe rin and ADAM15 were negatively related.Conclusions:1. ADAM l5 in local invasion and recurrence of breast cancer metastasis may participate in and played an important role. 2. ADAM l5 and E-cadherin in local infiltration and metastasis of breast cancer cells may exist mutual inhibition.
