The animals were randomly divided into six groups: NS group, NS+α group(phentolamine 10mg/kg), NS+β group(propranolol0.017mmol/mouse), Nes-1 group(100 pmol/mouse), Nes-1+α group and Nes-1+β group.Plasma FFA and insulin levels of each group were measured by ELISA. The protein and gene levels of AMPK /ACC were measured to reflect fatty acid oxidative. And the protein and gene levels of HSL/ATGL were detected to reflect lipid mobilization.Result: Central nesfatin-1 decreased the plasma FFA level and promotes the phosphorylation of HSL/ATGL in adipose tissue and AMPK/ACC in skeletal muscle when compared with the NS group mice. When blocked the α or β-adrenergic receptor used phentolamine and propranolol before nesfatin-1or NS treatment, the role of nesfatin-1 on plasma FFA, the phosphorylation of HSL/ATGL and AMPK/ACC were attenuated. The gene levels of HSL/ATGL and AMPK/ACC were altered with or without the presence of nesfatin-1 and the blocker, and the change of protein levels were consistent with the gene levels.Conclusions: Central nesfatin-1 stimulated fatty acid oxidation and promoted fat mobilization via sympathetic nervous system mediated AMP-activated protein kinase and HSL/ATGL pathway respectively. | | Keywords/Search Tags: | Nesfatin-1, type 2 diabetes mellitus, lipad metabolism, fat mobilization, sympathetic nervous system | PDF Full Text Request | Related items |
