Objective To explore the effects of high glucose on the expressions of urotensin II(UII) and its receptor(UT) in cardiac fibroblasts,to provide an experimental basis for further study of the role of UII system in diabetic cardiomyopathy.Methods The cardiac fibroblasts of aged 1-3 days old neonatal Wistar rats were obtained, cultured and passaged in vitro under sterile conditions.When the third-generation cardiac fibroblasts were cultured for 80% cell growth density should be evenly randomized to three groups: normal control,hypertonic control and high glucose. The three groups of cells were cultured with glucose 5.5mmol/L、glucose 5.5mmol/L + mannitol 19.5mmol/L and glucose 25.0mmol/L for 48 h respectively. Immumofluorescence and Western blotting were used to detect the expression of UII and UT.Results Immumofluorescence and Western blotting analysis showed that the expressions of UII and UT protein were significantly increased in high glucose group as compared with normal control and hypertonic control(F=15.08~192.2,q=3.751~30.00,P<0.05).Conclusion We found that UII and UT protein was expressed in rat cardiac fibroblasts and the expression of UII and UT can be stimulated by high glucose, which suggest UII system maybe involved in the process of diabetes myocardial fibrosis and provides a basis for further experiments. |
