Objective: This topic through shengyangyiwei Decoction of CAG rat model of intervention, observe the blocking atrophy gastritis rats of TLR4 Signal transduction pathway, to study the treatment mechanism of chronic atrophy gastritis, lay a preliminary basis for the further development of effective new drugs.Method:In the experiment, the physical factors of mould method CAG rat model was established, taking SPF adult Wister rats 90, weighing 200 ± 20 g, male and female 50 animals in each, giving the fed normally three days later, 10 rats were randomly selected,half male and half female, as the normal control group, the remaining 80 were 0.05%ammonia water, 10 mmol / L sodium deoxycholate, 30% alcohol and 2% salicylic acid mixture and immune method with disorder of hunger and satiety CAG rat model was established for 10 weeks.Build mode in the 7th week randomly 80 rats as model group,sucralfate divided into control group, low dose of shengyangyiwei Decoction intervention group, Shengyang Yiwei Decoction dose intervention group, high dose of shengyangyiwei Decoction intervention group, low dose of shengyangyiwei Decoction in the treatment group, Shengyang Yiwei Decoction dose treatment groups, of shengyangyiwei Decoction high dose treatment group, with 10 rats in each group(fmale =male= 5).After the success of the intervention in the intervention group after the success of the given control in model group and the dose group only modeling, sucralfate group was treated with sucralfate intragastric administration, the dose intervention group in modeling at the same time, with different doses of Shengyang Yiwei decoction treatment, after 4 weeks modeling, modeling the 10 week model group rats were sacrificed in gastric tissue biopsy to confirm modeling,in addition to the dose in the treatment group were treated with were killed to collect samples, the submission of the blank group, model group, sucralfate group, low dose,middle dose group, high dose intervention group rats gastric mucosa, the dose treatment group in modeling to continue treatment for 4 weeks after death, take out the stomach mucosa. Of the removed tissue using RT-PCR method and Western blot were used to detect TLR4, My D88, MAPK and NF kappa B and Ikappa B etc. index of gene and protein expression, and the income of the results were analyzed, in order to determine the effects of shengyangyiwei Decoction on rat CAG model of gastric mucosal tissue TLR4 Signal transduction pathway.Results:1.The successful establishment of rat CAG model.2.Expression of gene expression and protein of the indicators in the model group and blank control group have obvious difference, the difference has statistical significance(P <0.01), and accompanied by rat gastric mucosa atrophy, inflammatory cell infiltration. A few even accompanied with intestinal metaplasia and cancer precancerous lesions.3.Sucralfate in control group and model group compared to TLR4, My D88, MAPK and NF kappa B and Ikappa B gene expression and protein expression were significantly different(P < 0.01).4.Each dose Sun Yi Wei Decoction intervention group and treatment group of TLR4,My D88, MAPK and NF kappa B gene expression and protein expression gradually decreased with the increase of the concentration of and, and through the observation of the previous experimental observations and rat gastric mucosa can also see with Shengyang Yiwei Decoction concentration increases to high rats of the overall situation and the gastric mucosal histopathology were significantly improved.Conclusion:1.The CAG model of rats was successfully established by 10 weeks by using the physicochemical factors.2.My D88, MAPK, NF-, B kappa TLR4, I- kappa B and other indicators are the important signaling pathways of inflammatory reaction in chronic atrophic gastritis.3.Shengyangyiwei decoction has a blocking effect on the signal transduction pathway of TLR4 cells in atrophic gastritis rats.
