Objective:To investigate the expression of EGR-1 in normal ovarian tissues,benign ovarian tumour and malignant tumour,and to explore the correlation of EGR-1 with occurrence, development clinical significance in ovarian cancer.Method:1.A total of 20 samples of normal ovarian tissue, 20 samples of benign tumors and 60 cases of malignant tumors were collected.To choose these subjects based on postoperative pathological results of the first affiliated hospital of nanchang university and without any preoperative treatment.2.The expression of EGR-1 m RNA was detected by Real-time fluorescent quantitative PCR respectively in all collecting cases.3.The expression of EGR-1 protein was detected by immunohistochemical method respectively in all collecting cases.4. Follow up all the data of these subjects,included age,tumor type,lymphatic metastasis,tumorous grading and staging through telephone.Results:1.The expression of EGR-1 m RNAThe expression of EGR-1 m RNA,both in normal ovarian tissues(2.98±0.24)and benign ovarian tumour(2.87±0.20)were significant higher than ovarian cancer(1.02±0.16)(P 均<0.05). There were no significant differences between normal ovarian tissues and benign ovarian tumour(P >0.05).2.The expression of EGR-1 proteinImmunohistochemistry showed that positive expression of EGR-1 protein distribution in the nucleus,only a little expressed in cell cytoplasm.The expression of EGR-1 protein was judged by the cell staining intensity and positive percentage.The expression of EGR-1 protein,both in normal ovarian tissues(7.41±0.97)and benignovarian tumour(6.94±1.07)were significant higher than ovarian cancer(4.08±0.63)(P 均<0.05). There were no significant differences between normal ovarian tissues and benign ovarian tumour(P >0.05).3.The relationship between the expression of EGR-1 and the patients with ovarian cancer clinical indicatorsUsing statistical methods shows that the low expression of EGR-1 protein in ovarian cancer was correlated with surgical stages(χ2=7.500,P<0.05),degrees of pathological(χ2=8.612,P<0.05) and lymph node metastasis(χ2=5.714,P <0.05)but not with ages(χ2=0.782,P>0.05) and pathological types(χ2=0.717,P>0.05).4.Compare the five-year survival rate between the patient of ovarian cancerConsecutively followed up 5 years,34 cases of patients with low levels of Egr-1and 26 cases of patients with high levels,4cases lost folow-up.Based on the analysis of survival curves,the patients with low levels of Egr-1(32.4%)had a significantly lower five-year survival rate than that patients with a high levels of Egr-1(57.7%).Conclusion:The m RNA and protein levels of EGR-1 in ovarian malignant tumor significantly lower than that of the normal ovarian tissue and ovarian benign tumors.According to the correlation analysis that relationship between the expression of EGR-1 and the patients with ovarian cancer clinical indicators.Follow-up five year survival rate showed an the expression of relatively low survival rate EGR- 1was significantly lower than patients with relatively high expression of patients.Low expression of EGR-1 in ovarian malignant tumor play an important role in the occurrence and development of ovarian cancer, which is expected to conduct the grade, stage and prognosis judgement index of ovarian cancer.
