Objective:Myocardial infarction and other cardiovascular disease incidence increased year by year,the study found that the pathogenesis and myocardial ischemia reperfusion injury is closely related.Quercetin is a flavonoid plant estrogen,apple,red wine,onion,and other daily necessities were found in Green Tea.Studies have shown that[1,2],quercetin can protect against thrombosis,and formed by the reduction of oxygen free radicals,inhibition of apoptosis,mitochondrial membrane potential play a protective effect against myocardial ischemia reperfusion injury,but the specific mechanism is still not clear.In this experiment,H9C2 myocardial cells,construct hypoxia/reoxygenation?A/R?injury model,to further study the mechanism of quercetin on myocardial protective effect of hypoxia reoxygenation injury of oxygen,and the relationship between estrogen and estrogen ring finger protein?Efp?.Method:H9C2 cardiomyocytes with good morphology were selected and randomly divided into 5 groups after routine culture:?1?Control group;?2?A/R group;?3?Quercetin+A/R group;?4?Quercetin+negative Control+A/R group;?5?Quercetin+Efp-siRNA+A/R group.The cell viability was detected by MTS;Western blotting analysis assayed the expression of Efp;the flow cytometry was used to detect the degree of cell apoptosis,the intracellular ROS and the mitochondria membrane potential;Used the euzymelinked immunosorbent assay to analyze the content of Methane Dicarboxylic Aldehyde?MDA?and glutathione peroxidase?GSH-Px?and lactate dehydrogenase?LDH?,the activities of superoxide dismutase?SOD?.Results:1.Que pretreatment of myocardial cells of the expression level of Efp,compared with the level of hypoxia reoxygenation group Efp H9C2 expression,the expression level of Efp and pretreatment of quercetin dose dependently up-regulated?P<0.01?;2.Que pretreatment of myocardial cells of H9C2 group compared with hypoxia reoxygenation group,cell survival rate increased significantly,decreased the release of LDH?P<0.01?;3.Que pretreatment of myocardial cells of H9C2 group compared with hypoxia reoxygenation group,decreasing intracellular MDA,SOD and GSH-Px increased levels of antioxidant?P<0.01?;4.Que pretreatment of myocardial cells of H9C2 group compared with hypoxia reoxygenation group,intracellular ROS content decreased significantly,increase the stability of??m,degree of apoptosis decreased;5.Que pretreatment of myocardial cells of H9C2group after the addition of specific Efp-siRNA adenovirus,compared with hypoxia reoxygenation group,Que in myocardial hypoxia reoxygenation injury significantly reduced the protective effect of oxygen?P<0.01?.Conclusion:Quercein against myocardial anoxia-reoxygenation injury are involved in Estrogen-responsive finger protein pathway?Efp?,and at least partly Estrogen-responsive finger protein?Efp?.
