Objective:To investigate the influence of HER2 gene on the proliferation and invasion of IOMM-Lee maligant meningioma cell line, and to discuss whether the HER2 gene make the the proliferation and invasiveness effects by MAPK signal transduction pathway.Methods:1、 IOMM-Lee maligant meningioma cell lines were infected by silencing and overexpression of HER2 gene HER2 plamid with lentivirus vectors.The experiment was divided into five groups: the first group was untreated(blank),the second group was negative control of silence(NC-sh),the third group was the silence of HER2gene(HER2-sh),the fourth group was negative control of overexpressing(NC-ox),the fifth group was the overexpressing of HER2 gene(HER2-ox).The expression of HER-2 gene was evaluated by q-PCR and Western blot.2、 The MTT experiment, and EdU Labeling assay were to investgate the proliferation ability of different groups. Cell invasion and migration analysis were investgated by Matrigel invasion assay and Wound healing assay. Cell cycle and apoptosis assays by flow cytometry were to investgate the cell cycles and the ability of apotosis.3、 Western blot. were used to detect the expression of ERK1/2,P-ERK1/2,JNK,P-JNK,ERK5、P-ERK5、P38、P38 and ras protein.4、 The nude mice has been inocμlated with cells from each groups though their armpits, then, observe the time of forming and the weigh of tumors in the nude mice.Rsμlts:1、 the plasmid of silencing and the overexpression the HER2 were established successfμlly. The level of mRNA and protein of HER2 reached the requirements of each groups.2、 The proliferation, invasion and migration of the meningioma cells with a down-regμlation of HER-2 by shRNA humanly were significantly reduced. While the proliferation, invasion and migration of the human meningioma cells with a up-regμlation of HER-2 by shRNA were significantly enhanced. The influence of HER2 gene in the cell cycle was associated with G0/G1-phase to S-phase. The apoptosis of silencing HER2 group was increased and the apoptosis of overexpression HER2 group was decreased.3、 The influence of down-regμlation and up-regμlation the HER2 gene take effects on ERK1/2、P-ERK1/2、JNK、P-JNK、ERK5、P-ERK5、ras in MAPK signal pathway,but not on P38、P-P38.4、 Nude mice transplant tumor changes: the nude mice into tumor ability was abate, and the proliferation at a slower pace in the silencing group;On the contrary,the Overexpression of HER2 group,the tumor formation in nude mice was quick, and the invasion, migration were strengthen.Conclusions:1、 HER2 gene can promote the proliferation and invasion of malignant meningioma cells;2、 HER2 gene can take effects by the MAPK signaling pathways in malignant meningioma cells;...
