Objective:Through intraperitoneal injection of streptozocin to establishe diabetic rats model, and observe the effect of dexmedetomidine(DEX) on humoral immune funct-ion and complement component function, to investigate the role of dexmedetomidine on humoral immune function in diabetic rats.Method:Seventy-two male Sprague-Dawley rats, weighing 300-350 g,were randomly divided into 3 groups(n=24 each): control group(group C), diabetes mellitus group(group DM) and DEX + diabetes mellitus group(group DEX). The model of diabetes in group DM and group DEX was induced by intraperitioneal streptozocin60mg/kg and confirmed by blood glucose ≥16.7mmol/L,and underwent 30 min of exploratory laparotomy starting from 4 weeks after the diabetes model. Dexmedetom-idine 1μg/kg was infused iv over 10 min before operation, and then was infused at a rate of 5μg/kg·h maintain 4h in group DEX. Group C and group DM received the equal volume of normal saline. Six animals from each group selected before operation(T0), at the end of operation, 24 and 72 h after operation(T1~T3), the blood glucose was recorded, blood samples were obtained for determination of serum concentration of Ig A,Ig G,Ig M,CH50,C3 and C4. The body weight and weight of spleen were measure for spleen index.Result:(1) Compared with group C, the blood glucose concentration was increased at T0~3in group DM and group DEX(P<0.05). Compared with group DM, the blood glucose concentration was decreased at T1~3in group DEX(P<0.05),and there was no significant difference at T0.(2)Compared with group C, the serum concentration of Ig G and Ig M were decreased, the serum concentration of Ig A was increased at T0~3in group DM and group DEX(P<0.05). Compared with group DM, the serum concentration of Ig G and Ig M were increased, the serum concentration of Ig A was decreased at T1~3in groupDEX(P<0.05).(3)Compared with group C, the serum concentration of CH50,C3 and C4 was increased at T0~3in group DM and group DEX(P<0.05). Compared with group DM,the serum concentration of CH50,C3 and C4 was decreased at T1~3in group DEX(P<0.05).(4)Compared with group C, the spleen index were decreased at T0~3in group DM and group DEX(P<0.05). Compared with group DM, the spleen index were increased was decreased at T1~3in group DEX(P<0.05).Conclusion:Diabetes can induced the humoral immune function decreased and dysfunction of complement, humoral immunity and complement function can decline further after surgical trauma, and dexmedetomidine can improve the perioperative humoral immune function and complement component 3 in diabetic rats.
