Objective:To investigate the environment risk factors for breast cancer,and the relationship between seven SNPs,including 6788895(SIAH2 gene), rs10941679(5p12), rs889312(MAP3K1), rs13387042(2q35), rs6504950(17q23), rs17530068(6q14) andrs2284378(20q11) and female breast cancer in Ningxia.The study also detected the gene-environment interaction between seven SNPs and risk factors of breast cancer.Method:A hospital-based case-control study that the age as the frequency matching conditions was conducted in Affiliated General Hospital of Ningxia Medical University from May 2013 to July 2014.A total of 225 breast cancer patients and 225 controls enrolled in this study.Emerging female breast cancer in Ningxia as the case group,and the control group female come from the same hospital without breast cancer.Respondents needs to complete a same questionnaire,all information was collected by questionnaire,which included demographic data,menstrual history,reproductive history,lifestyle,family history of cancer and the other risk factors for breast cancer.we collected 2ml blood of respondents,and Sequenom Mass-ARRAY system was used to detect the genotype of the SNPs.SPSS 17.0 software was used for statistical analysis.Results:1.There was no significant different between case group and control group in marital status,education level,age,ethnic,occupational and stay up late(P>0.05).2.The age at first marriage,menstruation days,the age at first birth,the number of abortions,menopause,menstrual regularity and breast self-test were significant different between case group and control group(P<0.05).While no significant different was found in the age at menarche,menstrual cycle,the last of childbearing age,age at menopause,lactation age,passing through life,contraception,hospital breast examination,breast-feeding and BMI between the two groups(P>0.05).3.Physical exercise and wearing bar in evening were significant different between case group and control group(P<0.05).While no significant different was found in the smoking, passive smoking,drinking,insomnia,napping and eating between the two groups(P>0.05).4.Family history of cancer was significant different between case group and control group(P<0.05),and the proportion of family history of cancer populations in the case group(9.8%) was higher than that in the control group(1.8%).5.Logistic regression analysis indicated that the number of abortions(OR=1.565) and family history of cancer(OR=5.030) were the risk factors of breast cancer,breast self-text(OR=0.418),physical exercise(OR=0.444) and at night not wearing bars(OR=0.562) were the protect factors of breast cancer.6.The genotype and allele distribution of the seven SNPs(rs6788895, rs10941679, rs889312、rs13387042、rs6504950、rs17530068 and rs2284378)were not significantly different between cases and controls.7.There were no interactions between SNPs(rs6788895, rs889312, rs13387042, rs6504950, rs17530068,rs10941679 and rs2284378) and the four environment factors(the number of abortions, family history of cancer, breast self-test,physical exercise,at night not wearing bars) of breast cancer(P>0.05).Conclusions:1The number of abortions,breast self-test,the physical exercise,wearing bar in evening and family history of cancer might be the environment factors for the female breast cancer.2.The seven SNPs(rs6788895, rs10941679, rs889312, rs13387042, rs6504950, rs17530068 and rs2284378) may not relate with the breast cancer.3.There were no interactions between SNPs(rs6788895, rs889312, rs13387042, rs6504950, rs17530068,rs10941679 and rs2284378) and environment factors of breast cancer.
