Objective Combining Cyclin D1 and PCNA to investigate the expression and possible mechanism of CREPT in renal clear cell carcinoma(RCCC) corresponding noncancerous tissues.Method Real-time PCR and western blot were used to investigate expression of m RNA and protein level of CREPT, Cyclin D1 and PCNA in 40 specimens of RCCC and corresponding noncancerous tissues; To explore the correlation between CREPT and Cyclin D1 and CREPT and PCNA in RCCC. Immunohistochemical method was adopted to examine protein expression o f CREPT in 105 specimens of RCCC and corresponding noncancerous tissues to investigate its expression and clinical significance in RCCC.Results Real-time PCR suggested that the m RNA level of CREPT,Cyclin D1 and PCNA in RCCC were much higher than corresponding noncancerous tissues(( 3.034±1.868 vs1.000±0.000, P <0.05;5.611±4.089 vs 1.000±0.000,P <0.05;7.527±6.3801.000±0.000,P <0.05); Western blot revealed that protein level of CREPT(1.062±0.090 vs 0.618±0.139,P=0.000),Cyclin D1(1.066±0.165 vs 0.712±0.165,P=0.001<0.05)and PCNA(1.033±0.091 vs 0.715±0.144,P=0.003<0.05) in RCCC were highly expressed and in corresponding noncancerous tissues were lowly. The expression of m RNA and protein were positive correlation between CREPT and Cyclin D1 in RCCC(r=0.875, P=0.000;r=0.786,p=0.000). The expression of CREPT and PCNA in m RNA and protein level of RCCC were no correlation(r=0.673,P=0.37>0.05; r=0.589,P=0.74>0.05). Immunohistochemistry revealed that CREPT in RCCC of the positive rate was 79%, in the corresponding noncancerous tissues positive rate was 44.8%. A statistically significance of CREPT was in RCCC andcorresponding noncancerous tissues(P<0.05). The level of CREPT protein expression did not correlate with gender,age, lymph node metastasis and clinic stag tumor size(P>0.05). The level of CREPT protein expression correlated with histological grade(P<0.05).Conclusion CREPT is overexpressed in RCCC, and it may be a useful tool in the diagnosis of RCCC as well as new target treatment. The expression of m RNA and protein were positive correlation between CREPT and Cyclin D1 and no correlation between CREPT and PCNA suggesting that the possible mechanism of CREPT promotes tumorigenesis of RCCC by interacting with Cyclin D1.
