Objective : Analysis the effect of Dextran sulfate to the expression of Hif-1α,VEGF,MVD and the number of Intra-abdominal tumor nodules, to research the mechanism of Dextran sulfate inhibited the human gastric cancer cells celiac planting metastasis.Method: The 72 nude mice were divided into experimental group(40) and control group(40). The experimental group injected BGC-823 human gastric cancer cells 0.2 m L into abdominal cavity at a concentration of 1 × 107 / m L, after 24 h injected 0.3% DS1 m L into abdominal cavity.The control group injected the same concentration of cells 0.2 m L into abdominal cavity, after 24 h injected saline 1m L into abdominal cavity. When 1th,3th,7th,14 th days killed the mouse by cervical dislocation method.Observe the number and size of tumor nodules within the laparotomy,the expression of Hif-1α, VEGF, CD34 by immunohistochemistry and expression of Hif-1α, VEGF m RNA and protein by RT-PCR and Western- blot.Result: 1 One day after human gastric cancer cells were injected into abdominal cavity, the tumor nodules were not observed in the control group and the experimental group,over time, the number of tumor nodules have increased in the two groups, the rate of increase in the control group was significantly higher than the experimental group.and the number of nodules in control group and the experimental group were different significantly(P <0.001).2. Immunohistochemical results showed that: the expression of Hif-1α at 1th day was no different in the two groups(P> 0.05), 3th,7th,14 th days the expression of Hif-1α were increased and were significantly different(P <0.01); the expression of VEGF in the control group and the experimental group were increased and the two groups were significantly different(P <0.001), the experimental group was significantly lower than the control group; early MVD value was significantly increased over the time, but at a later stage the MVD value declines, the MVD values of the control group and the experimental group in different points were significantly different(P <0.01), the MVD values of the experimental group was significantly lower than the control group.3. The RT-PCR results showed that: Over time, the expression of the Hif-1α m RNA, and the VEGF m RNA in the control group and the experimental group were increased and were significantly different(P <0.001), the experimental group was significantly lower than the control group.4. The western-blot results showed that:the 1th day the expression of the Hif-1α and the VEGF in the control group and the experimental group was not detected after human gastric cancer cells were growed, over time,the expression of the Hif-1α and VEGF are in two groups were increased and were significantly different(P <0.001), the experimental group were significantly higher than the control group. 5. Over time, the expression of the Hif-1α and the VEGF in the control group and the experimental group was increased, and the expression of the Hif-1α and the VEGF were positive correlation.Conclusion: 1. DS can inhibit the metastasis of human gastric cancer cells into the peritoneal cavity. 2. Over the time, the expression of the Hif-1α, and the VEGF in the control group and the experimental group were increased; the expression of the Hif-1α, VEGF and the MVD values were significantly different, and the experimental group was significantly lower than the control group.3. The expression of the Hif-1α and the VEGF 、the VEGF and the MVD values were positive correlation. 4. DS inhibited the peritoneal metastasis of human gastric cancer cells mighe by down-regulating the expression of Hif-1α, VEGF and MVD.
