Trametes versicolor is one kind of medicinal macrofungi and is used widely in modern times. In this paper, we made a series of herbal textual research on T. versicolor and ‘Qingzhi’. At the same time, the betulinic acid was isolated and purified from T. versicolor. In order to fully use it, we also studied on the pharmacological activity and pharmacokinetic in rats.In the part of textual research, we firstly studied on the designation, morphological characteristics, geographical distribution and medicinal efficacy of “Qingzhi” in ancient time. Then a comparative research was done between Qingzhi and T. versicolor, one of modern traditional Chinese medicine(TCM), on their morphology, habitat and geographical distribution. According to the characteristic of Qingzhi in the ancient literature of herbal medicine and combining with the modern pharmacological activity research of T. versicolor, we could made the conclusion that the ancient Qingzhi and T. versicolor is the same species of Polyporaceae.In the study of T. versicolor extraction, we focus on the extraction and purification of betulinic acid. Then the betulinic acid was further evidenced and highly purified by HPLC, which is for the next study of a series of pharmacological activity. The extraction process of betulinic acid is that the powder of fruiting bodies was extracted by heating method with 75% ethanol and extracted with petroleum ether. Separation and purification were used with the silica gel column chromatography and gel layer chromatography, ultimately, get the compound of betulinic acid( Purity ≥95%).The therapeutic effect of T. versicolor fruiting bodies extract and betulinic acid from the extract on improving the gastrointestinal function of mice, and the effect of betulinic acid on lipid peroxidation in the mice(P<0.01) were studied. The gastrointestinal function injury of mice was modeled by rhubarb and sennoside decoction. The results indicated that T. versicolor extract and betulinic acid can improve the behavior of the mice, increasing the secretory levels of gastrin, pepsin, gastric acid and decreasing the level of blood MDA. The fruiting bodies extract can improve the activity of blood Se-GSH-Px, SOD and BAT as compared with the control group and model group(P<0.01), and the effect of betulinic acid on anti-lipid-peroxidation of the mice is one of the treatment function.The protective effects of T. versicolor extract for acetaminophen acute liver damage. One-hundred and twenty mice were randomly divided into twelve groups: blank group, model group and some treatment groups. Mice liver injury was induced by periton injection paracetamol physiologic saline solution at a dose of 600mg/ml. The activities of serum alanine am inotransferase(ALT) and aspirate am inotransferase(AST), the contents of glutathione(GSH) in the liver injury. And the pathologic changes of liver were observed. The results of T. versicolor extracts except Ethyl acetate extracts significantly decreased the activities of ALT and AST in serum(P<0.05 or P<0.01) and increased the GSH content in liver tissue(P<0.05 or P<0.01), and improved liver histopathological changes. T. versicolor is effective in protection against acetaminophen-induced liver damage.In this paper we study on pharmacokinetics in vivo of rats used by the HPLC, then verified by HPLC-MS. As the result of that the metabolism of betulinic acid was quickly absorbed and distributed in rat blood after oral administration. Calculated by 3P97 using compartmental analysis that the distribution processes could be fitted to a two-compartment model. Oral administration performance volume of distribution(V/ F) showed that betulinic acid can be rapidly distributed in the extravascular system. And clearance(CL/F) data showed that betulinic acid had a faster rate of excretion. In tissue metabolism, compound contents were measured in 0.5h, 1.5h, 3h three times and the results showed that tissues contain the compound and a high content in the lung, 2.24μg/ml in 0.5h. The content is higher in the spleen, but relatively lower in the brain and heart. After 3h, the contents in the organizations were basically down to a similar level. In the feces and urine excretion rate test, the urine contained less betulinic acid content, but higher content in feces, which can be drawn in betulinic acid is mainly excreted through the feces.The acquired data would provide helpful and scientific information about the T. versicolor and betulinic acid for development and utilization of medicine and health food.
